目的研究重组人血管内皮抑制素与白细胞介素-2治疗卵巢癌的疗效。方法将80只卵巢癌裸鼠模型随机分为4组阴性对照组、阳性对照组及实验A、B组,每组各20只。阴性对照组未作特殊处理;阳性对照组予以尾静脉注射10 mg·kg~(-1)重组人血管内皮抑制素注射液,qd;实验A组予以肌内注射白细胞介素-2 1.0×10~6U·d~(-1),qd;实验B组予以转染白细胞介素-2基因的羊水干细胞,尾静脉注射0.2 m L微泡化,qd。4组疗程均为7周。每隔3 d测量一次肿瘤体积,末次处理后5 d剖取瘤块测体积、重量,计算抑瘤率。用TUNEL检测肿瘤细胞凋亡情况。结果 4组的肿瘤体积随时间逐渐增加,阴性对照组增加趋势明显高于阳性对照组和实验A、B组(P<0.05),但阳性对照组和实验A、B组比较差异无统计学意义(P>0.05)。治疗后,阳性对照组和实验B组的肿瘤重量分别为(878.3±30.5),(902.4±34.6)mg明显低于阴性对照组和实验A组的肿瘤重量(1554.7±45.3),(1484.6±41.4)mg(P<0.05),且阳性对照组的肿瘤重量和实验B组比较、阴性对照组的肿瘤重量和实验A组比较,差异无统计学意义(P>0.05)。阳性对照组和实验B组的抑瘤率和肿瘤凋亡细胞数均明显高于阴性对照组和实验A组(P<0.05),且阳性对照组和实验B组的抑瘤率和肿瘤凋亡细胞数比较、阴性对照组的抑瘤率和肿瘤凋亡细胞数和实验A组比较,差异无统计学意义(P>0.05)。结论转染白细胞介素-2基因的羊水干细胞可以抑制卵巢癌实体瘤的生长,促进肿瘤细胞凋亡。
Objective To study the therapeutic effect of recombinant human endostatin and interleukin-2 on ovarian cancer. Methods 80 ovarian cancer nude mice model were randomly divided into 4 negative control group, positive control group and experimental A, B group, each group of 20. Negative control group without special treatment; positive control group was given intravenous injection of 10 mg · kg -1 recombinant human endostatin injection, qd; experimental group A intramuscular injection of interleukin -2 1.0 × 10 ~ 6U · d ~ (-1), qd. In experimental group B, transfused IL-2 gene-derived amniotic fluid stem cells were injected into the tail vein with 0.2 mL microbubbles, qd. 4 groups of treatment are 7 weeks. The tumor volume was measured every 3 days. The volume and weight of tumor mass were measured 5 days after the last treatment, and the tumor inhibition rate was calculated. TUNEL detection of tumor cell apoptosis. Results The volume of tumor in 4 groups increased gradually with time and the increasing trend in negative control group was significantly higher than that in positive control group and experimental group A and B (P <0.05), but there was no significant difference between positive control group and experimental group A and B (P> 0.05). After treatment, the tumor weights of the positive control group and experimental group B were (878.3 ± 30.5) and (902.4 ± 34.6) mg, respectively, which were significantly lower than those of the negative control group and experimental group A (1554.7 ± 45.3) and (1484.6 ± 41.4) ) mg (P <0.05). The tumor weight of the positive control group was not significantly different from that of the experimental group B (P> 0.05). The tumor inhibition rate and the number of apoptotic cells in positive control group and experimental group B were significantly higher than those in negative control group and experimental group A (P <0.05), and the tumor inhibition rate and tumor apoptosis in positive control group and experimental group B There was no significant difference between the control group and the control group (P> 0.05). Conclusion Amniotic fluid stem cells transfected with interleukin-2 gene can inhibit the growth of solid tumor in ovarian cancer and promote the apoptosis of tumor cells.
