目的:探讨血清miR-19a和miR-19b诊断胃癌的价值及其与临床病理特征的关系。方法:采用实时荧光定量PCR(RT-q PCR)技术检测61例正常人和61例胃癌患者血清中miR-19a和miR-19b的表达量,分析其表达与胃癌临床病理参数的关系,并绘制ROC曲线分析其诊断胃癌的价值。结果:胃癌患者血清miR-19a和miR-19b相对表达水平均高于正常对照组(P<0.05),其表达水平与临床分期及淋巴结有无转移显著相关(P<0.05),而与性别、年龄及吸烟史无相关性(P>0.05);miR-19a的ROC曲线下面积0.989,灵敏度95.1%,特异度94.0%;miR-19b的ROC曲线下面积0.983,灵敏度93.4%,特异度94.0%。结论:血清中miR-19a和miR-19b可作为一种非损伤性诊断方法,辅助胃癌诊断。
Objective: To investigate the value of serum miR-19a and miR-19b in the diagnosis of gastric cancer and its relationship with clinicopathological features. METHODS: The expression of miR-19a and miR-19b in serum of 61 normal and 61 gastric cancer patients was detected by real-time RT-PCR (RT-q PCR). The relationship between the expression of miR-19a and miR-19b and the clinicopathological parameters of gastric cancer was analyzed. ROC curve analysis of its value in the diagnosis of gastric cancer. Results: The relative expression levels of serum miR-19a and miR-19b in patients with gastric cancer were higher than those in the normal control group (P<0.05). The expression levels of miR-19a and miR-19b were significantly associated with clinical stage and lymph node metastasis (P<0.05). There was no correlation between age and smoking history (P>0.05). The area under the ROC curve of miR-19a was 0.989, the sensitivity was 95.1%, the specificity was 94.0%; the area under the ROC curve of miR-19b was 0.983, the sensitivity was 93.4%, and the specificity was 94.0% . Conclusion: Serum miR-19a and miR-19b can be used as a non-invasive diagnostic method to assist in the diagnosis of gastric cancer.
