目的建立森林脑炎、登革热和乙型脑炎病毒液相蛋白芯片检测方法。方法将病毒抗原包被微球,利用病毒抗体和阳性血清建立并优化森林脑炎、登革热和乙型脑炎病毒液相蛋白芯片检测方法,评价方法的敏感性和准确性。结果建立的液相蛋白芯片检测方法病毒抗体和生物素标记二抗的孵育时间均为1 h,生物素标记羊抗兔Ig G和羊抗人Ig G最佳稀释度分别为1∶200和1∶100,能对3种病毒单一和混合感染准确检测。对295份临床样本的检测结果表明,该方法具有高通量、快速、敏感、特异的特点。结论本方法能对3种虫媒病毒同时进行快速检测,为虫媒病毒的筛查检测提供一种新方法。
Objective To establish a method for detection of forest encephalitis, dengue fever and Japanese encephalitis virus liquid protein chip. Methods The virus antigens were coated on microspheres, and virus encephalitis and positive sera were used to establish and optimize forest encephalitis, dengue fever and encephalitis virus liquid protein chip detection methods to evaluate the sensitivity and accuracy of the method. Results The detection method of liquid protein chip established the incubation time of virus antibody and biotin labeled secondary antibody for 1 h, and the optimal dilutions of biotin labeled goat anti-rabbit Ig G and goat anti-human Ig G were 1: 200 and 1 : 100, which can accurately detect single and mixed infections of 3 kinds of viruses. The test results of 295 clinical samples show that the method has high-throughput, rapid, sensitive and specific characteristics. Conclusion The method can simultaneously detect three kinds of arbovirus simultaneously and provide a new method for the screening and detection of the arbovirus.
