目的:观察中药复方洁泽1号(JZ1)与有效避孕剂量的外用杀精剂壬苯醇醚(N-9)的复合制剂洁泽2号(JZ2)对小鼠滴虫性阴道炎的预防作用。方法:将112只小鼠随机分为JZ2组(高、中、低)、JZ1组(高、中、低)、壬苯醇醚(N-9)组、甲硝唑组、空白凝胶组及模型组。造模前,每只小鼠阴道给予相应的药物凝胶或生理盐水20μl,注药2min后每只小鼠接种滴虫液20μl,在接种后的2、4、7、10、12、14天取小鼠阴道冲洗液涂片镜检及培养,观察小鼠感染情况,并计算各组小鼠不同时间的感染率。结果:JZ2组、N-9组小鼠感染率都为0,与相同时间模型组比较差异有显著性意义(P<0.01);甲硝唑组小鼠感染率很低,前4天仅为10%,之后为0,前10天与模型组比较有显著性差异(P<0.01);JZ1组小鼠滴虫感染率比模型组低,2g.ml-1浓度组在第2天与模型组比较差异有显著性意义(P<0.05);空白凝胶组小鼠在不同时间的滴虫感染率与模型组比较差异均无显著性意义(P>0.05);JZ2组、N-9组、JZ12g.ml-1和1g.ml-1剂量组小鼠滴虫感染率与甲硝唑组比较差异无显著性意义(P>0.05);JZ2与JZ1组比较,0.5g.ml-1组在第2天感染率差异有显著性(P<0.05)。结论:预防性给药后,JZ2高、中、低剂量组和N-9组都能完全预防小鼠滴虫性阴道炎的感染,其作用强于同浓度的JZ1。
OBJECTIVE: To observe the prevention of trichomonal vaginitis in mice by JZ2, a compound preparation of traditional Chinese medicine compound JZ1 (JZ1) and an effective contraceptive dose of external spermicidal agent nonoxynol (N-9) effect. METHODS: One hundred and twelve mice were randomly divided into JZ2 group (high, middle, low), JZ1 group (high, middle, low), nonoxynol (N-9) group, metronidazole group, blank gel group. And model groups. Before the modeling, 20 μl of the corresponding drug gel or physiological saline was given to each mouse in the vagina, and 20 μl of trichomonas liquid was inoculated to each mouse 2 minutes after the injection, which was 2, 4, 7, 10, 12, and 14 days after inoculation. Smears and cultures of mice’s vaginal washing solution were taken for microscopy and culture. The infection status of the mice was observed, and the infection rate of each group of mice at different times was calculated. Results: The infection rates of mice in the JZ2 and N-9 groups were all 0, which was significantly different from the model group at the same time (P<0.01). The rate of infection in the metronidazole group was very low, only in the first 4 days. 10%, then 0. The difference between the first 10 days and the model group was significant (P<0.01); the infection rate of Trichomonas in the JZ1 group was lower than that in the model group, and the 2g.ml-1 group was on the 2nd day with the model. There was significant difference between the groups (P<0.05). There was no significant difference in the infection rates of the trichomonas between the blank gel group and the model group (P>0.05). The JZ2 group and the N-9 group. There was no significant difference in the infection rate of trichomoniasis between the JZ12g.ml-1 and 1g.ml-1 dose groups and the metronidazole group (P>0.05). The JZ2 and the JZ1 group were compared with the 0.5g.ml-1 group. There was a significant difference in the infection rate on the second day (P<0.05). Conclusion: After prophylactic administration, the JZ2 high, middle, low dose group and N-9 group can completely prevent the infection of trichomonal vaginitis in mice. Its effect is stronger than the same concentration of JZ1.
