目的:比较化痰祛瘀、益气养阴、疏肝理气3种不同方剂对兔动脉内膜损伤后血管平滑肌细胞(VSMCs)增殖的影响。方法:将46只新西兰兔随机分为正常组、模型组、氟伐他汀对照组、化痰祛瘀组、益气养阴组、疏肝理气组,正常组6只,其余每组各8只。通过高脂饲料配合腹主动脉内膜剥脱术建立兔动脉内膜损伤模型。取腹主动脉下段,电镜观察血管平滑肌细胞形态;应用RT-PCR法测定原癌基因c-myc和血小板源性生长因子-BB(PDGF)水平;免疫组化法测定胰岛素样生长因子-1(IGF-1)水平。结果:(1)模型组PDGF、c-myc、IGF-1均显著高于空白组(P<0.05)。(2)氟伐他汀组、化痰祛瘀组、益气养阴组、疏肝理气组与模型组相比较PDGF、c-myc、IGF-1显著下降(P<0.05)。(3)化痰祛瘀组、益气养阴组与疏肝理气组比较PDGF、IGF-1显著下降(P<0.05),c-myc差异不显著(P>0.05)。结论:(1)动脉损伤后可致PDGF、c-myc、IGF-1表达增加,提示PDGF、c-myc、IGF-1表达增加在动脉损伤后平滑肌细胞增殖中起一定作用。(2)化痰祛瘀、益气养阴、疏肝理气3种方剂均能使动脉损伤后PDGF、c-myc、IGF-1表达减少,对动脉损伤后平滑肌细胞增殖有一定抑制作用,化痰祛瘀与益气养阴方效果更显著。
OBJECTIVE: To compare the effects of three different prescriptions of Huatan, Qi and Yin, and Shugan Liqi on the proliferation of vascular smooth muscle cells (VSMCs) after intimal injury in rabbits. METHODS: Forty-six New Zealand rabbits were randomly divided into normal group, model group, fluvastatin control group, Huatan group, Yiqi Yangyin group, Shugan Liqi group, 6 normal rats, and 8 rats in each group. . A rabbit model of arterial intimal injury was established by high fat diet with abdominal aortic endarterectomy. The lower abdominal aorta was harvested and the morphology of vascular smooth muscle cells was observed by electron microscopy. The levels of proto-oncogene c-myc and platelet-derived growth factor-BB (PDGF) were measured by RT-PCR, and insulin-like growth factor-1 was measured by immunohistochemistry. IGF-1) levels. Results: (1) PDGF, c-myc and IGF-1 in the model group were significantly higher than those in the blank group (P<0.05). (2) PDGF, c-myc and IGF-1 were significantly decreased in fluvastatin group, phlegm-removing group, Yiqiyangyin group, Shuganliqi group compared with the model group (P<0.05). (3) The levels of PDGF and IGF-1 were significantly decreased (P<0.05), and the difference of c-myc was not significant (P>0.05) between the phlegm-resolving group, the Yiqi Yangyin group and the Shugan Liqi group. Conclusion: (1) The expression of PDGF, c-myc and IGF-1 can be increased after arterial injury, which suggests that the increased expression of PDGF, c-myc and IGF-1 plays a role in the proliferation of smooth muscle cells after arterial injury. (2) The three kinds of prescriptions can reduce the expression of PDGF, c-myc and IGF-1 after arterial injury, and inhibit the proliferation of smooth muscle cells after arterial injury. The effect of wolfberry and Qi replenishing yin is more significant.
