目的:探讨不同浓度的胰岛素对高糖条件下人脐静脉内皮细胞(human umbilical vein endothelial cells,HUVEC)凋亡及小窝蛋白(caveolin,Cav)-1表达的影响。方法:体外培养HUVEC,分为A组(对照组,葡萄糖浓度5.5 mmol/L)、B组(高糖组,葡萄糖浓度33.3 mmol/L)、C组(高糖+低浓度胰岛素组,葡萄糖浓度33.3 mmol/+胰岛素浓度20 mU/L)、D组(高糖+高浓度胰岛素组,葡萄糖浓度33.3 mmol/L+胰岛素浓度1 000 mU/L)。采用流式细胞仪检测细胞凋亡比例及细胞周期,免疫组化检测HUVEC的Cav-1表达。结果:在高糖条件下,HUVEC凋亡比例显著升高,停滞于G0/G1期细胞增加,Cav-1表达减少;低浓度胰岛素使细胞凋亡比例减少,Cav-1表达增加;加入高浓度胰岛素不能改变细胞凋亡比例及细胞周期,Cav-1表达亦无明显改变。结论:低浓度胰岛素可抑制高糖导致的内皮细胞凋亡,而高浓度胰岛素无此作用,其机制可能与Cav-1表达有关。
Objective: To investigate the effects of different concentrations of insulin on the apoptosis of human umbilical vein endothelial cells (HUVECs) and the expression of caveolin (Cav) -1 under high glucose conditions. Methods: HUVECs were cultured in vitro and divided into group A (control group, glucose concentration 5.5 mmol / L), group B (high glucose group, glucose 33.3 mmol / L), group C (high glucose + low concentration insulin group, glucose concentration 33.3 mmol / L insulin concentration 20 mU / L), D group (high glucose + high concentration insulin group, glucose concentration 33.3 mmol / L + insulin concentration 1000 mU / L). Flow cytometry was used to detect the percentage of apoptosis and cell cycle. The expression of Cav-1 in HUVECs was detected by immunohistochemistry. Results: The apoptosis rate of HUVEC was significantly increased under high glucose conditions. The cells arrested in G0 / G1 phase increased and the expression of Cav-1 decreased. Low concentration of insulin decreased the percentage of apoptosis and increased the expression of Cav-1. Insulin can not change the proportion of apoptosis and cell cycle, Cav-1 expression also did not change significantly. CONCLUSION: Low concentration of insulin can inhibit endothelial cell apoptosis induced by high glucose, while high concentration of insulin has no effect, which may be related to the expression of Cav-1.
