目的研究全反式维甲酸(ATRA)对人肝细胞癌细胞BEL7404细胞生长的影响。方法藉MTT法观察ATRA(10-5mol/L)对BEL7404细胞的生长及其对人表皮生长因子刺激的反应性的影响。结果ATRA可使BEL7404细胞生长抑制,10-5mol/LATRA作用5天,细胞生长抑制率为60%。经ATRA预处理5天后的细胞,换不含ATRA的培养液,在第5天生长抑制率为22%(P<0.01),且其对人表皮生长因子促增殖作用的反应性显著降低,表皮生长因子作用的细胞增殖率,对照组为177.9%±1.0%(72h),ATRA预处理组为162.5%±2.3%(72h)(P<0.01)。结论ATRA可能藉降低细胞对表皮生长因子刺激的反应性抑制人肝细胞癌细胞的增殖。
Objective To investigate the effect of all-trans retinoic acid (ATRA) on the growth of human hepatocellular carcinoma cell line BEL7404. Methods MTT assay was used to observe the effect of ATRA (10-5 mol/L) on the growth of BEL7404 cells and its response to human epidermal growth factor stimulation. RESULTS: ATRA inhibited the growth of BEL7404 cells, and the effect of 10-5 mol/LATRA for 5 days showed a 60% inhibition of cell growth. After 5 days of ATRA-pretreatment, cells without ATRA had a growth inhibition rate of 22% on the 5th day (P<0.01), and their reactivity to human epidermal growth factor was significantly reduced. The proliferation rate of cells treated with epidermal growth factor was 177.9%±1.0% (72 hours) in the control group and 162.5%±2.3% (72 hours) in the ATRA pretreatment group (P<0.01). . Conclusion ATRA may inhibit the proliferation of human hepatocellular carcinoma cells by reducing the response of cells to epidermal growth factor stimulation.
