目的观察左归复方(滋阴益气活血解毒组方)干预治疗MKR小鼠后糖代谢、血清胰岛素及胰岛B细胞胰岛素表达的变化。方法40只MKR小鼠经鉴定后,随机分为模型组,左归复方高、低剂量组,阳性对照组(文迪雅),每组10只,另设10只C57小鼠为野生鼠组。各药物组分别以相应剂量连续给药30d,模型组和野生鼠组灌胃蒸馏水。于给药前、第15日及第30日分别作空腹血糖测定;末次给药后0.5h心脏取血,放射免疫法测血清胰岛素,免疫组织化学SABC法检测胰岛B细胞胰岛素表达。结果左归复方干预治疗后,其高剂量具有显著的降低MKR小鼠空腹血糖、改善高胰岛素血症的作用。MKR小鼠胰岛B细胞胰岛素表达显著高于C57野生型小鼠;左归复方高剂量组干预治疗后胰岛B细胞胰岛素表达显著下调;左归复方高剂量组下调胰岛B细胞胰岛素表达与对照药文迪雅作用相当。结论左归复方具有改善MKR小鼠糖代谢、减轻胰岛B细胞的工作负荷、减少胰岛素表达及分泌的作用。
Objective To observe the changes of glycometabolism, serum insulin and insulin expression of islet B cells after intervention of Zuoguifufang (ZIYIN YIQI HUOXUEJUEDUJUFANG) in the treatment of MKR mice. Methods After 40 MKR mice were identified, they were randomly divided into model group, Zuogui Fufang high- and low-dose group, positive control group (Avandia), 10 in each group, and another 10 C57 mice in wild mice group. . Each drug group was continuously administered with the corresponding dose for 30 days. The model group and the wild mice group were given distilled water. Fasting blood glucose was measured before administration, on the 15th and 30th day; blood was taken from the heart 0.5 h after the last administration, serum insulin was measured by radioimmunoassay, and insulin expression was detected by immunohistochemistry SABC method. Results After intervention therapy of Zuogui Fufang, its high dose significantly reduced fasting blood glucose and improved hyperinsulinemia in MKR mice. The insulin expression of islet B cells in MKR mice was significantly higher than that of C57 wild mice; the expression of insulin in islet B cells was significantly down-regulated in the high-dose group of Zuogui compound; the insulin expression of islet B cells in high-dose group of Zuogui compound was reduced and compared Diya is quite effective. Conclusion Zuoguifufang can improve the glucose metabolism of MKR mice, reduce the workload of islet B cells, and reduce the insulin expression and secretion.
