目的:考察中药中黄酮、酚酸类成分清除2,2′-azobis(2-methylpropionamidine)dihydrochloride(AAPH)的活性及构效关系,中药提取物清除AAPH的活性与其酚类成分之间的相关性。方法:以中药中常见的22种黄酮、酚酸类化合物和9种中药为研究对象,以Oxygen Radical Absorbance Capacity(ORAC)法对上述31个样品清除AAPH自由基的活性进行评估,以Folin-Ciocalteu法测定中药中黄酮和酚酸类成分的总量,以高效液相色谱-紫外检测技术对中药的抗氧化活性成分进行定性定量检测。结果:茶叶清除AAPH自由基能力最强,ORAC值为4 786.40μmol.g-1;红花醇提物最弱,ORAC值784.04μmol.g-1。有效成分中,槲皮素清除AAPH自由基能力最强,ORAC值为12.90μmol.g-1;(-)-EGC最弱,ORAC值2.47μmol.g-1。中药清除AAPH自由基能力(Y)与总黄酮和酚酸类成分含量(X)之间存在相关性,Y=1 844.8Ln(X)-3 577.5,r=0.867 5。结论:9种中药清除AAPH自由基的有效成分为黄酮和酚酸类成分,这些化合物清除AAPH自由基的能力与其立体构型、是否糖苷化及酯化等因素有关。将ORAC法与高效液相色谱法相结合,对中药抗氧化活性及有效成分进行研究,是一个切实可行的方法。
OBJECTIVE: To investigate the activity and structure-activity relationship of 2,2’-azobis (2-methylpropionamidine) dihydrochloride (AAPH) extracted from flavonoids and phenolic acids in traditional Chinese medicine (TCM). The correlation between the activity of AAPH and the phenolics . Methods: Twenty-two kinds of flavonoids, phenolic acids and nine kinds of traditional Chinese herbs were studied in this study. The activity of AAPH free radical scavenging was evaluated by Oxygen Radical Absorbance Capacity (ORAC). Folin-Ciocalteu Method to determine the total amount of flavonoids and phenolic acids in traditional Chinese medicine, qualitative and quantitative detection of the antioxidant activity of traditional Chinese medicine by high performance liquid chromatography - ultraviolet detection technology. Results: Tea had the strongest ability to scavenge AAPH free radicals with a ORAC value of 4 786.40 μmol.g-1. The weakest alcohol extract of Safflower was ORAC value of 784.04μmol.g-1. Active ingredient, quercetin scavenging AAPH free radical ability, ORAC value of 12.90μmol.g-1; (-) - EGC weakest, ORAC value of 2.47μmol.g-1. There was a correlation between AAPH free radical scavenging capacity (Y) and content of total flavonoids and phenolic acids (X), Y = 1844.8Ln (X) -3 577.5, r = 0.867 5. CONCLUSION: The active components of AAPH free radical scavenging by 9 Chinese herbs are flavonoids and phenolic acids. The ability of these compounds to scavenge AAPH free radicals is related to their stereo configuration, glycosylation and esterification. Combining ORAC with HPLC, it is a feasible method to study the antioxidant activity and active ingredients of Chinese traditional medicine.
