目的 探讨用温度灭活的SARS冠状病毒作为免疫用抗原的可行性。方法 采用细胞培养、酶联免疫吸附试验 (ELISA)对灭活前后SARS冠状病毒感染性和抗原性进行检测。结果 5 6℃、30min对SARS冠状病毒感染性的灭活率为 99 99% ;5 6℃、6 0min对SARS冠状病毒感染性的灭活率为 1 0 0 .0 0 % ;检测病毒灭活前、灭活 30min、灭活 6 0min抗原性P/N值分别为 5 4 4、3 0 2、2 72。结论 随着灭活时间的延长 ,病毒的抗原性降低 ,5 6℃、6 0min可能可以作为SARS冠状病毒灭活的一种方法
Objective To investigate the feasibility of using temperature-inactivated SARS coronavirus as antigen for immunization. Methods The infection and antigenicity of SARS coronavirus before and after inactivation were detected by cell culture and enzyme-linked immunosorbent assay (ELISA). Results The inactivation rate of SARS coronavirus was 99 99% at 5 6 ℃ and 30 min. The inactivation rate of SARS coronavirus at 56 ℃ and 60 min was 100.0% Before antigen inactivation 30min, inactivated 6min antigenic P / N values were 544,3 0 2,2 72. Conclusion With the prolongation of inactivation time, the antigenicity of the virus is reduced. At 60 ℃ and 60 min, it may be used as a method to inactivate SARS coronavirus
