目的:研究N-马来酰-L-缬氨酸酯姜黄素(MVC)对胃癌MGC-803细胞周期的作用机制。方法:采用MTT法检测MVC对肿瘤细胞的生长抑制作用;采用流式细胞术检测MVC对MGC-803细胞周期的影响;采用Western blot印迹法检测MVC对MGC-803细胞周期相关基因蛋白p21WAF1/CIP1,Cdc 2和Cyclin B1表达的影响。结果:20μmol.L-1 MVC可明显抑制MGC-803细胞生长,可明显诱导MGC-803细胞S,G/M期阻滞。20μmol.L-1MCV作用MGC-803细胞24 h,p21WAF1/CIP1蛋白表达增高,Cdc 2蛋白表达减少。结论:MVC可通过增加细胞周期相关基因p21WAF1/CIP1蛋白的表达和下调Cdc 2蛋白表达,使MGC-803细胞的周期阻断在S,G2/M期,从而抑制MGC-803细胞的生长。
AIM: To investigate the mechanism of action of N-maleyl-L-valine ester curcumin (MVC) on gastric cancer MGC-803 cell cycle. Methods: MTT assay was used to detect the inhibitory effect of MVC on tumor cells. Flow cytometry was used to detect the effect of MVC on the cell cycle of MGC-803 cells. Western blotting was used to detect the effect of MVC on the expression of p21WAF1 / CIP1 , Cdc 2 and Cyclin B1 expression. Results: 20μmol.L-1 MVC could significantly inhibit the growth of MGC-803 cells and significantly induce the arrest of S phase and G / M phase in MGC-803 cells. After treated with 20μmol·L-1MCV for 24 hours, the expression of p21WAF1 / CIP1 protein and the expression of Cdc2 protein decreased in MGC-803 cells. CONCLUSION: MVC can inhibit the growth of MGC-803 cells by increasing the expression of p21WAF1 / CIP1 and down-regulating the expression of Cdc 2, and blocking the cycle of MGC-803 cells in S and G2 / M phases.
