玫瑰花形成法通常被使用于分离T和B淋巴细胞。因为所形成的玫瑰花不稳定,我们用AET处理SRBC以增强SRBC和T淋巴细胞的结合能力,然后用密度梯度离心将T和B细胞分离。 由SRBC-AET所形成的玫瑰花和没有用AET处理的SRBC所形成的玫瑰花相比较具有形成率高,更加稳定和更好的重复性。 所回收细胞悬液中活细胞量用染料排除试验超过90%。T和B细胞纯度用免疫荧光试验来估价分别为80%和70%。因此,所得到的B细胞可以被用于HLA-DR分型。本文对国产AET和进口AET的效应也进行了比较。
Rose formation is commonly used to isolate T and B lymphocytes. Because of the instability of the roses formed, we treated SRBC with AET to enhance the binding capacity of SRBC and T lymphocytes, and then T and B cells were separated by density gradient centrifugation. Roses formed from SRBC-AET had higher rate of formation, more stability and better repeatability than roses formed from ATR-treated SRBC. The amount of viable cells in the recovered cell suspension was over 90% with a dye exclusion test. The purity of T and B cells was estimated by immunofluorescence to be 80% and 70%, respectively. Therefore, the resulting B cells can be used for HLA-DR typing. This article also compares the effects of domestic AETs and imported AETs.
