目的检测YAP在人结直肠腺癌中的表达,探讨其与肿瘤细胞增殖、凋亡的关系及意义。方法应用免疫组织化学Polymer conjugate方法对10例正常结直肠组织、10例癌旁组织、20例结直肠息肉、40例结直肠腺瘤和80例结直肠腺癌组织进行YAP检测,并采用DNA末端原位标记染色法(TUNEL)检测肿瘤组织的细胞凋亡情况。结果免疫组织化学结果显示YAP主要在结直肠腺上皮组织的细胞核中表达。10例正常结直肠组织YAP阳性表达率为0,10例癌旁组织为0,80例结直肠腺癌组织中YAP的表达阳性率为63%,而结直肠息肉和腺瘤的YAP表达阳性率分别为5%、33%。5组间的差异有统计学意义(P<0.05)。在结直肠腺瘤组中,轻、中、重度非典型增生3组YAP表达之间的差异有统计学意义(P<0.05)。YAP的表达与结直肠腺癌的分化程度之间的差异有统计学意义(P<0.05),而与年龄、性别、肿瘤大小等之间比较差异无统计学意义。结直肠腺癌组织中YAP的表达水平与凋亡率呈负相关,差异有统计学意义(P<0.05)。结论 YAP参与结直肠腺癌的发生发展,可以作为此病的早期诊断指标。
Objective To detect the expression of YAP in human colorectal adenocarcinoma and to explore its relationship with the proliferation and apoptosis of tumor cells. Methods Tissue samples from 10 cases of normal colorectal tissue, 10 cases of paracancerous tissues, 20 cases of colorectal polyps, 40 cases of colorectal adenomas and 80 cases of colorectal adenocarcinoma were detected by YAG, TUNEL method was used to detect the apoptosis of tumor tissue. Results Immunohistochemistry showed that YAP was mainly expressed in the nucleus of colorectal glandular epithelium. The positive expression rates of YAP in 10 cases of normal colorectal tissues were 0, 10 cases of paracancerous tissues were 0, and in 80 cases of colorectal adenocarcinoma tissues, the positive rate of YAP expression was 63%, while the positive rate of YAP expression in colorectal polyps and adenomas Respectively 5%, 33%. The difference between the 5 groups was statistically significant (P <0.05). In colorectal adenoma group, the difference of YAP expression in three groups of mild, moderate and severe atypical hyperplasia was statistically significant (P <0.05). There was a significant difference between the expression of YAP and the differentiation of colorectal adenocarcinoma (P <0.05), but not with the age, gender, tumor size and so on. The expression level of YAP in colorectal adenocarcinoma was negatively correlated with the apoptosis rate, the difference was statistically significant (P <0.05). Conclusion YAP is involved in the development of colorectal adenocarcinoma and can be used as an early diagnostic indicator of the disease.
