目的观察早期糖尿病大鼠肾脏肌球蛋白轻链激酶(MLCK)表达及意义。方法腹腔注射链尿佐菌素(65mg/kg)诱发糖尿病模型,应用胰岛素控制血糖,心脏取血检测血糖及肾功能相关指标。通过免疫组化与Western-blot检测肾组织肌球蛋白轻链激酶表达。结果与正常对照组相比,模型组血糖及肾功能相关指标发生显著性变化。正常组肾小球、肾小管及肾脏血管均有MLCK的表达。模型组与正常对照组相比,肾小球表达MLCK有一定的减弱,治疗组与正常对照组相比无明显变化。结论早期糖尿病肾病形成可能是由于肌球蛋白轻链激酶表达降低所致。胰岛素治疗糖尿病肾病机制可能是增加肾脏肌球蛋白轻链激酶的表达。
Objective To observe the expression of myosin light chain kinase (MLCK) in early diabetic rats and its significance. Methods Diabetic rats were induced by intraperitoneal injection of streptozotocin (65mg / kg), insulin was used to control blood glucose, and blood was taken from the heart to detect blood glucose and renal function. The expression of myosin light chain kinase in renal tissue was detected by immunohistochemistry and Western-blot. Results Compared with the normal control group, the indexes of blood glucose and renal function in the model group changed significantly. The normal group glomerular, renal tubular and renal vascular MLCK expression. Compared with the normal control group, the expression of MLCK in glomerulus decreased to some extent in the model group, and there was no significant change in the treatment group compared with the normal control group. Conclusion The formation of early diabetic nephropathy may be due to the decreased expression of myosin light chain kinase. Insulin treatment of diabetic nephropathy may be increased renal myosin light chain kinase expression.
