目的研究大孔树脂纯化马兰总三萜的工艺。方法以齐墩果酸为对照品,以吸附量和解吸率作为效果评定指标,筛选出对马兰中总三萜纯化效果最好的大孔树脂,并对上样量、上样液浓度、洗脱速率、洗脱乙醇浓度及用量参数进行考察,筛选出最佳工艺条件。结果 HPD-500的吸附量为0.495 5mg/mL,解吸率为30.47%,效果最佳;最佳上样液浓度:0.02g/mL;流速:2mL/min;洗脱剂:70%乙醇;洗脱体积:2倍柱体积。纯化后马兰三萜含量可达到17.81%。结论 HPD-500大孔树脂在所得工艺条件下可以对马兰中总三萜进行较好的纯化。
Objective To study the process of purifying total triterpenoids from Malan by macroporous resin. Methods Oleanolic acid was used as a reference substance, adsorption and desorption rates were taken as the evaluation indexes of effect, and the macroporous resin with the best purifying effect on the total triterpenoids in Malan was screened. The effects of the amount of sample, concentration of the sample solution, Off rate, elution ethanol concentration and dosage parameters were investigated, the best screening process conditions. Results The adsorption capacity of HPD-500 was 0.495 5 mg / mL and the desorption rate was 30.47%. The optimum concentration of the sample was 0.02 g / mL. The flow rate was 2 mL / min. The eluent was 70% ethanol. Volume off: 2 column volumes. After purification Malan triterpene content can reach 17.81%. Conclusions HPD-500 macroporous resin can be used to purify the total triterpenoids from Malan.
