目的探讨软肝冲剂对四氯化碳肝硬化大鼠P2 1mRNA表达及细胞周期各时相DNA含量的影响 ,从mRNA、DNA水平探讨软肝冲剂阻止肝硬化的作用机理。方法采用RT PCR及流式细胞计数仪分别检测四氯化碳肝硬化大鼠P2 1mRNA表达、DNA含量。结果软肝冲剂能降低四氯化碳肝硬化大鼠P2 1mRNA表达 ,与模型组比较有显著性差异 (P <0 .0 1 ) ,大剂量组的疗效与小剂量组比较无明显差异 (P >0 .0 5 )。正常肝组织P2 1mRNA呈相当低水平表达 ,其表达相对值为 (0 .1 7± 0 .0 6)U。定量DNA图像分析发现模型组中处于G1期的细胞数要明显多于其它各组 (P <0 .0 1 ) ,软肝冲剂大剂量组中处于G1期的细胞数要明显少于软肝冲剂小剂量组 (均P <0 .0 1 )。结论软肝冲剂能调节P2 1基因表达 ,调节细胞周期 ,促进肝细胞再生 ,抑制肝纤维化、肝硬化的形成。
Objective To investigate the effect of Ruangan Granules on P2 1 mRNA expression and cell cycle DNA content in CCl4 induced liver cirrhosis rats, and to explore the mechanism of Ruangan Granules in preventing liver cirrhosis from mRNA and DNA levels. Methods RT-PCR and flow cytometry were used to detect P2 1 mRNA expression and DNA content in CCl4 cirrhosis rats. Results Ruangan Granules can reduce the expression of P2 1 mRNA in CCl4-induced liver cirrhosis rats, which is significantly different from that in the model group (P < 0.01). The efficacy of high-dose group has no significant difference compared with the low-dose group (P <0.01). >0 .0 5 ). The normal liver tissue P2 1 mRNA was expressed at a relatively low level, and the relative expression value was (0.17 ± 0.66) U. Quantitative DNA image analysis showed that the number of cells in the G1 phase in the model group was significantly higher than that in the other groups (P < 0.01). The number of cells in the G1 phase in the high-dose soft liver granule group was significantly less than that in the soft liver granules. Small dose group (all P <0.01). Conclusion Ruangan Granules can regulate the expression of P2 1 gene, regulate cell cycle, promote hepatocyte regeneration, and inhibit the formation of hepatic fibrosis and cirrhosis.
