目的探讨组蛋白去乙酰化酶3(histone deacetylase 3,HDAC3)在肝细胞癌(hepatocellular carcinoma,HCC)组织中的表达及其临床意义.方法采用免疫组织化学法检测60例HCC组织及癌旁组织石蜡切片中HDAC3的表达,分析其与HCC临床病理特征及与患者预后的关系.结果 HCC组织中HDAC3的表达水平明显高于其相应癌旁组织(P<0.05).HDAC3在HCC中的表达水平与HCC患者的性别、年龄、乙型肝炎病毒感染、TNM分期、病理分级、AFP水平、是否合并肝硬化、肿瘤数目、肿瘤大小和肿瘤结节的单发多发数目等无关(P>0.05),与肿瘤复发有相关性(P<0.05).HDAC3的表达与p-STAT3的表达呈正相关(r~2=0.622,P<0.001).HDAC3高表达组患者的术后生存时间明显低于低表达组患者(P<0.05),提示HDAC3高表达可能与HCC患者的预后有关.结论 HDAC3表达升高可能与HCC的发生、发展密切相关.
Objective To investigate the expression of histone deacetylase 3 (HDAC3) in hepatocellular carcinoma (HCC) and its clinical significance. Methods Immunohistochemistry was used to detect 60 cases of HCC and adjacent tissues. The expression of HDAC3 in paraffin sections was analyzed to analyze the relationship between the expression of HDAC3 and clinicopathological characteristics of HCC and the prognosis of HCC. The expression level of HDAC3 in HCC tissues was significantly higher than that in corresponding paracancerous tissues (P<0.05). HDAC3 expression level in HCC It was not related to gender, age, hepatitis B virus infection, TNM staging, pathological grade, AFP level, whether or not complications of liver cirrhosis, number of tumors, tumor size and number of single tumors in HCC patients (P>0.05). There was a correlation with tumor recurrence (P<0.05). The expression of HDAC3 was positively correlated with the expression of p-STAT3 (r~2=0.622, P<0.001). The survival time of HDAC3 high expression group was significantly lower than that of low expression. Patients (P<0.05) suggested that the high expression of HDAC3 may be related to the prognosis of patients with HCC. Conclusion The increased expression of HDAC3 may be closely related to the occurrence and development of HCC.
