研究喜树碱(Cam)或和环胞菌素A(CyA)合用能否诱导抗三尖杉酯碱的人白血病HL60细胞(HR20)凋亡.采用细胞计数法、染色质凝集观察、流式细胞术和DNA琼脂糖凝胶电泳进行研究.Cam诱导敏感HL60细胞凋亡;作用HR20细胞24h,仅将细胞阻断于G2期;0.2,1mg·L-1Cam作用48h后,部分细胞发生凋亡.1mg·L-1CyA明显增加柔红霉素在HR20细胞内的积聚,但并不增加Cam诱导的凋亡细胞数量.结果表明:Cam诱导HR20细胞发生细胞凋亡需较长的时间,其作用不受CyA的影响.
To investigate whether the combination of camptothecin (Cam) or cyclosporin A (CyA) can induce apoptosis of HL60 cells (HR20) against human harringtonine. Cell counting, chromatin agglutination, flow cytometry and DNA agarose gel electrophoresis were used for the study. Cam induced apoptosis of sensitive HL60 cells; HR20 cells treated with HR20 cells for 24 h only blocked cells in G2 phase; 0.2 and 1 mg·L-1 Cam treated for 48 h, apoptosis occurred in some cells. 1 mg·L-1 CyA significantly increased redness. The accumulation of ADM in HR20 cells did not increase the number of apoptotic cells induced by Cam. The results showed that the induction of apoptosis in HR20 cells by Cam takes a long time, and its effect is not affected by CyA.
