目的:研究芪贞固本方对Lewis肺癌小鼠的转移抑制作用及作用机制。方法:建立Lewis肺癌小鼠模型,按实验分组干预,计算比较各组抑瘤率、肺转移抑制率,免疫组化法检测E-钙黏素表达,RT-PCR法检测VEGFmRNA表达。结果:芪贞固本方有一定的抑瘤及肺转移抑制作用,抑瘤率及肺转移抑制率分别为:13.97%、19.13%;与对照组相比,芪贞固本方可明显下调VEGFmRNA的表达,促进E-钙黏素的表达(P<0.05),与顺铂合用时作用增强。结论:芪贞固本方可抑制Lewis肺癌小鼠转移,其机制可能是促进E-钙黏素的表达,下调VEGFmRNA的表达。
Objective: To study the inhibitory effect of Qizhen Guben Fang on Lewis lung carcinoma in mice and its mechanism. Methods: Lewis lung cancer mice model was established. The intervention groups were divided into experimental groups. The inhibition rate of tumor and the rate of pulmonary metastasis were calculated and compared. The expression of E-cadherin was detected by immunohistochemistry. The expression of VEGF mRNA was detected by RT-PCR. Results: Qizhen Guben Fang had certain anti-tumor and lung metastasis inhibitory effects, inhibition rate and lung metastasis inhibition rate were: 13.97%, 19.13%; compared with the control group, Qizu Guben significantly decreased VEGF mRNA The expression of E-cadherin (P <0.05) increased with the combination of cisplatin. Conclusion: Qizhen Guben can inhibit the metastasis of Lewis lung cancer in mice, the mechanism may be to promote the expression of E-cadherin, down regulate the expression of VEGFmRNA.
