目的研究促凋亡基因PDCD5和Caspase-3 mRNA在多发性骨髓瘤(MM)中的表达,并讨论其在MM的发生、发展及预后中的意义。方法从收集40例MM患者及10例正常对照的骨髓标本中取骨髓单个核细胞(BM-MCs),采用实时荧光定量PCR(Real ti me PCR)的方法检测BMMCs中PDCD5和Caspase-3 mRNA的表达,同时收取血浆以检测β2微球蛋白(β2-MG)分泌水平,Durie-Sal mon(D-S)分期标准对MM患者进行临床分期。结果与对照组比较,PDCD5和Caspase-3 mRNA在MM组中表达明显减低(P<0.05),差异有统计学意义;复发/难治组患者PDCD5和Caspase-3 mRNA的表达仍保持较低水平;初发组与复发/难治组之间表达差异无统计学意义。PD-CD5和Caspase-3 mRNA表达与患者病程D-S分期和β2-MG(≥或<4.0 g/L)水平有负相关性(r=-0.86,P<0.05)。结论MM患者PDCD5和Caspase-3 mRNA表达水平降低,提示PDCD5和Caspase-3可能参与MM的发生发展,可能作为评判MM患者预后的新指标。
Objective To study the expression of pro-apoptotic genes PDCD5 and Caspase-3 mRNA in multiple myeloma (MM) and discuss its significance in the occurrence, development and prognosis of MM. Methods Bone marrow mononuclear cells (BM-MCs) were obtained from bone marrow samples collected from 40 patients with MM and 10 normal controls. Real-time quantitative PCR (Real-time PCR) was used to detect the expression of PDCD5 and Caspase-3 mRNA in BMMCs The plasma was collected to detect the level of β2 microglobulin (β2-MG) secretion. The patients were divided into clinical stage and Durie-Sal mon (DS) stage. Results Compared with the control group, the expression of PDCD5 and Caspase-3 mRNA in MM group was significantly lower (P <0.05), the difference was statistically significant; the expression of PDCD5 and Caspase-3 mRNA in patients with relapsed / refractory group remained low There was no significant difference in the expression between the primary group and the recurrent / refractory group. There was a negative correlation between the expression of PD-CD5 and Caspase-3 mRNA in patients with D-S stage and β2-MG (≥ or <4.0 g / L) (r = -0.86, P <0.05). Conclusion The decreased expression of PDCD5 and Caspase-3 mRNA in MM patients suggests that PDCD5 and Caspase-3 may play a role in the development of MM, which may be used as a new index to evaluate the prognosis of MM patients.
