目的 :检测氢氧化钙置于根管不同位置根尖周pH值变化及牙髓卟啉单胞菌(Porphynomonasl endodontics,P.e)刺激成骨细胞IL-6、TNF-α表达的影响。方法:选取140颗无菌人单根管离体牙,根管预备后随机分为6个实验组(每组20个样本)和1个对照组(20个样本)。1~3组分别将手调氢氧化钙糊剂封入根管下1/2、根管上1/2和髓室,4~6组将Apexcal封入根管下1/2、根管上1/2和髓室,对照组不封药。每组随机选取10颗离体牙置于P.e悬液中,于封药后3、7 d测量各组根尖周区细菌对MC3T3-E1细胞IL-6、TNF-α表达的影响。每组剩余10颗离体牙置于蒸馏水中,检测封药后3、7、14、21 d根尖周pH值变化。采用SPSS 13.0软件包对数据进行统计学分析。结果:2种氢氧化钙封入根管不同位置后,随着时间延长,根尖周pH值升高;14 d时达到高峰。手调氢氧化钙糊剂充填髓室组根尖周pH值升高程度与其他实验组相比有显著差异,根尖周P.e菌液预处理的MC3T3-E1细胞IL-6、TNF-α表达量较对照组显著降低,各实验组间IL-6、TNF-α表达量无显著差异。结论:氢氧化钙糊剂封入根管不同部位均使根尖周pH升高,抑制根尖周P.e刺激成骨细胞表达IL-6、TNF-α。
PURPOSE: To investigate the effects of calcium hydroxide on the apical pH and the expression of IL-6 and TNF-α in osteoblasts stimulated by Porphyromonas endodontics (P.e). Methods: One hundred and forty sterile single root canal teeth were selected and divided into 6 experimental groups (20 samples per group) and 1 control group (20 samples) after root canal preparation. 1 to 3 groups were hand-adjusted calcium hydroxide paste sealed root canal 1/2, root canal 1/2 and medullary, 4 to 6 group Apexcal sealed root canal 1/2, 1 / 2 and pulp chamber, the control group is not sealed drugs. Each group of 10 randomly selected teeth were placed in P.e suspension at 3,7 d after sealant to measure each group of periapical area bacteria in MC3T3-E1 cells IL-6, TNF-α expression. The remaining 10 teeth in each group were placed in distilled water, and the changes of apical pH value at 3, 7, 14, and 21 days after sealant were measured. Data were statistically analyzed using SPSS 13.0 software package. Results: After the two kinds of Ca (OH) 2 were encapsulated in different positions of root canal, the pH value of periapical period increased with time and peaked at 14 days. Compared with other experimental groups, the pH value of periapical pH value in hand-adjusted calcium hydroxide paste-filled pulp chamber group was significantly different. The expression of IL-6 and TNF-α in MC3T3-E1 cells Compared with the control group, the levels of IL-6 and TNF-α in the experimental groups were significantly decreased. CONCLUSION: Calcium hydroxide paste encapsulated in different parts of root canal can increase the pH of periapical tip and inhibit the expression of IL-6 and TNF-α in osteoblasts stimulated by P.e.
