【目的】观察柴甘解忧汤对帕金森抑郁(PDD)大鼠海马神经元及脑源性神经生长因子(BDNF)表达的影响。【方法】选用成年雄性SD大鼠24只,随机分为4组:正常组、模型组、柴甘解忧汤组、氟西汀组,每组6只,除正常组外,其他组大鼠均单笼饲养。帕金森抑郁模型复制成功后,模型组、柴甘解忧汤组、氟西汀组分别以蒸馏水2.0 m L、柴甘解忧汤(剂量为8 g·kg~(-1)·d~(-1))及氟西汀药液(剂量为1.8 mg·kg~(-1)·d~(-1))灌胃,连续21 d。采用尼氏染色及免疫组织化学法观察海马神经元损伤情况及BDNF表达的改变。【结果】与正常组比较,模型组大鼠海马神经元出现明显的形态学损伤;柴甘解忧汤、氟西汀均可改善PDD大鼠海马神经元的损伤,对海马神经元细胞有一定的保护作用。与正常组比较,模型组大鼠海马区BDNF阳性细胞数与阳性染色面积、积分光密度、平均光密度均显著下降(P<0.01)。与模型组比较,柴甘解忧汤组、氟西汀组海马BDNF阳性细胞数与阳性染色面积、积分光密度、平均光密度均显著上升(P<0.05或P<0.01)。【结论】柴甘解忧汤能减轻帕金森抑郁大鼠海马神经元损伤和增加BDNF阳性表达,这可能是其抗帕金森抑郁的作用机理之一。
【Objective】 To observe the effect of Chagan Jiexiu decoction on the expression of hippocampal neurons and brain-derived nerve growth factor (BDNF) in Parkinson’s Depression (PDD) rats. 【Methods】 Twenty-four adult male Sprague-Dawley rats were randomly divided into 4 groups: normal group, model group, Chagangan Jieyu Decoction group and fluoxetine group, with 6 rats in each group. Except normal group, Single cage feeding. After the successful replication of Parkinson’s depression model, the rats in model group, Chaigan Jieyu Decoction group and fluoxetine group were treated with 2.0 m L of distilled water, Chaigan Jieyu Decoction (dosage of 8 g · kg -1 · d -1 ) And fluoxetine (1.8 mg · kg -1 · d -1) for 21 days. The damage of hippocampal neurons and the change of BDNF expression were observed by Nissl staining and immunohistochemistry. [Results] Compared with the normal group, the hippocampal neurons of the model group showed obvious morphological damage. Chai Gan Jie Yu Tang and fluoxetine could improve the damage of hippocampal neurons in PDD rats, Protective effects. Compared with the normal group, the number of BDNF positive cells, the positive staining area, the integral optical density and the average optical density of hippocampus in model group decreased significantly (P <0.01). Compared with model group, BDNF positive cells, positive staining area, integral optical density and average optical density in hippocampus of Chagangan Jieyu Decoction and fluoxetine group were significantly increased (P <0.05 or P <0.01). 【Conclusion】 Chaganan Jieyu Decoction can reduce the damage of hippocampal neurons and increase the expression of BDNF in the hippocampus of the rats with Parkinsonism, which may be one of its anti-Parkinson’s depression mechanisms.
