[目的]研究尖萼耧斗菜外植体消毒及愈伤组织的诱导。[方法]采用不同消毒剂种类和浓度,对尖萼耧斗菜种子进行消毒处理,研究其污染率和污染速率,寻找建立无菌苗培养系最适宜的方法;以尖萼耧斗菜种子获得的无菌苗的根、茎、叶片为外植体,通过在MS基本培养基中附加不同激素配比,筛选最佳外植体和最佳培养基配方。[结果]外植体材料最适宜的灭菌处理方法为:先将外植体在75%酒精中浸30s,用无菌水冲洗5遍,再将其用0.2%升汞液浸泡2min,再用无菌水冲洗5遍。该消毒方法使污染率最低、发芽率最高,并且获得的无菌苗长势最好;诱导尖萼耧斗菜胚性愈伤组织的最佳外植体为幼根;诱导胚性愈伤组织的最佳培养基为MS0.6mg/L2,4-D+0.5mg/L6-BA。[结论]该研究为尖萼耧斗菜的大规模生产提供了技术支持,并为其药用和观赏价值的最大发挥作出了贡献。
[Objective] The research aimed to study the disinfection of callus and callus induction of Calyx. [Method] Different types and concentrations of disinfectants were used to sterilize the seeds of Calyx dicotyledon, to study their pollution rate and pollution rate, and to find the most suitable method to establish aseptic seedling culture system. The roots, stems and leaves of sterile seedlings were used as explants. The optimal explants and the best medium formula were screened by adding different hormones in MS basal medium. The disinfection method has the lowest pollution rate and the highest germination rate, and the best sterile seedling is obtained. The best explant for inducing embryogenic callus of Cephalopoda sp. Is young root; the induction of embryogenic callus The best medium was MS0.6mg / L2,4-D + 0.5mg / L6-BA. [Conclusion] The study provided technical support for the large-scale production of Calyx, and contributed to its maximum medicinal value and ornamental value.
