目的 观察体外培养中亚硒酸钠的浓度变化和培养时间的延长对Bcap 37人乳腺癌细胞增殖及酶活性的影响。 方法 在体外培养Bcap 37人乳腺癌细胞中加入不同浓度的亚硒酸钠 ,常规染色显示其形态和增殖情况及各组Bcap 37人乳腺癌细胞内的酸性磷酸酶、碱性磷酸酶和非特异性酯酶活性。 结果 2 μmol L的亚硒酸钠对肿瘤细胞的生长具有一定的抑制作用 ,而 10 μmol L、5 0 μmol L的亚硒酸钠具有一定的细胞毒性。Bcap 37人乳腺癌细胞内的酸性磷酸酶、碱性磷酸酶、非特异性酯酶活性随着硒浓度的加大 ,培养时间的延长呈下降趋势 ,并且10 μmol L、5 0 μmol L亚硒酸钠培养 2d后 ,3种酶的活性完全消失。 结论 硒对肿瘤细胞的生长和酶的活性都具有抑制作用 ,并且两者之间存在着一致性。
Objective To observe the effects of concentration of sodium selenite and prolongation of culture time on the proliferation and enzyme activity of Bcap 37 human breast cancer cells in vitro. Methods Different concentrations of sodium selenite were added into Bcap 37 human breast cancer cells in vitro. The morphology and proliferation of Bcap 37 human breast cancer cells were observed by routine staining. The contents of acid phosphatase, alkaline phosphatase and nonspecific Esterase activity. Results Sodium selenite (2 μmol L) had a certain inhibitory effect on the growth of tumor cells. However, 10 μmol L and 50 μmol L sodium selenite had certain cytotoxicity. The activities of acid phosphatase, alkaline phosphatase and non-specific esterase in Bcap 37 human breast cancer cells decreased with the increase of selenium concentration, and the prolongation of incubation time showed a decreasing trend. Moreover, 10 μmol L, 50 μmol L selenious acid After 2 days of sodium culture, the activities of the three enzymes completely disappeared. Conclusion Selenium has an inhibitory effect on tumor cell growth and enzyme activity, and there is consistency between the two.
