目的 研究血源α 干扰素与基因工程α 干扰素对大鼠肝脏贮脂细胞增殖和胶原合成的影响 ,为临床应用基因工程α 干扰素抗纤维化提供参考。方法 分离、培养大鼠贮脂细胞 ,采用MTT法和3H 脯氨酸参入法分别测定细胞增殖和胶原合成。结果 血源α 干扰素与基因工程α 干扰素在1× 10 6 ~ 1× 10 7U·L-1浓度范围内均能显著抑制大鼠肝脏贮脂细胞增殖和胶原合成 ,二者间相比差异无显著性。结论 血源α 干扰素与基因工程α 干扰素均有良好的体外抗肝纤维化作用 ,且作用相似。
Objective To study the effect of interferon-alpha (IFN-α) and genetically engineered interferon-alpha (IFN-α) on the proliferation and collagen synthesis of rat liver fat-storing cells, and provide a reference for the clinical application of genetically engineered IFN-α for anti-fibrosis. Methods The lipid-storing cells were isolated and cultured in vitro. Cell proliferation and collagen synthesis were determined by MTT assay and 3H-proline incorporation assay. Results Both IFNα and IFNα in genetically engineered interferon alpha could significantly inhibit the proliferation and collagen synthesis of rat fat cells in the concentration range of 1 × 10 6 to 1 × 10 7 U · L -1 No significant. Conclusion Both interferon-alpha and gene-interferon alpha have good anti-hepatic fibrosis effect in vitro with similar effects.
