目的研究49个不同产地的牛蒡种质资源的千粒质量、发芽势、发芽率及萌发初期的牛蒡苷与苷元的积累动态。方法在培养箱中进行种子发芽实验。HPLC法测定萌发种芽中牛蒡苷与苷元的量,色谱柱为AgilentTC-C18(250mm×4.6mm,5μm);流动相:甲醇-水(线性梯度洗脱:0min,水55%;10min,水46%;20min,水30%);体积流量:1.0mL/min;检测波长:280nm。结果成熟饱满的牛蒡种子千粒质量、发芽势、发芽率高,萌发初期的牛蒡苷的量下降,牛蒡苷元的量升高。结论本实验建立的方法为牛蒡子药材的质量评价提供参考。
Objective To study the accumulation of arcarythin and aglycone in 1000 germplasm resources, germination potential, germination rate and burdock burdock germplasm resources in 49 burdock germplasm resources of different origins. Methods Seed germination experiments were carried out in an incubator. The amount of arctiin and aglycone in germination seed was determined by HPLC. The column was Agilent TC-C18 (250 mm × 4.6 mm, 5 μm). The mobile phase was methanol-water (linear gradient elution: 0 min, water 55% Water 46%; 20min, water 30%); volumetric flow rate: 1.0mL / min; detection wavelength: 280nm. Results The mature and full-burdock seed had the highest quality of 1000-grain weight, germination potential and germination rate. The amount of arctiin decreased at the early stage of germination, and the amount of arctigenin increased. Conclusion The method established in this experiment provides a reference for the quality evaluation of Arctii.
