目的预测乙型肝炎病毒e抗原(HBeAg)和c抗原(HBcAg)的B细胞表位。方法用Lasergene软件包中的Protean软件和吴玉章的氨基酸抗原指数预测法对HBeAg和HBcAg的氨基酸序列进行分析,预测其B细胞表位。结果推测HBeAg最有可能的B细胞表位位于其N端第50~63、137~146、85~94区段和第150~157区段内或它们的附近,但其他区域如HBeAg N-端第37~43、12~18区段和第24~32区段内或它们的附近也可能存在B细胞表位;HBcAg最有可能的B细胞表位位于其N-端第40~53、127~136、150~181、75~84区段和第140~147区段内或它们的附近,但其他区域如HBcAg N-端第27~33、2~8区段和第14~22区段内或它们的附近也可能存在B细胞表位。结论用多种方法预测HBeAg和HBcAg的B细胞表位,并对其表位进行比较分析,为制备更好的HBV感染者血清学检测相关诊断用品的研究提供线索。
Objective To predict the B cell epitopes of hepatitis B virus e antigen (HBeAg) and c antigen (HBcAg). Methods The amino acid sequences of HBeAg and HBcAg were predicted by Protease software of Lasergene and amino acid antigen index prediction of Wu Yuzhang to predict their B cell epitopes. As a result, it was speculated that the most probable B cell epitopes of HBeAg were located in or near the N-terminal 50th to 63th, 137th to 146th, 85th to 94th and 150th to 157th sections, but the other regions such as HBeAg N-terminal B cell epitopes may also be present in or around the 37th to 43th, 12th to 18th, and 24th to 32nd segments; the most probable B cell epitopes of HBcAg are located at N-terminal 40th to 53th, 127th ~ 136, 150 ~ 181, 75 ~ 84 and 140 ~ 147 segments or in the vicinity thereof, but other regions such as the 27 ~ 33,2 ~ 8 segments and the 14 ~ 22 segments of the HBcAg N- B cell epitopes may also be present within or adjacent to them. Conclusions The B cell epitopes of HBeAg and HBcAg are predicted by a variety of methods and their epitopes are comparatively analyzed. This study may provide clues for the study of serological detection related diagnostic products for better HBV infection.
