目的:探讨运动对大鼠心肌营养因子1(cardiotrophin-1,CT-1)以及信号转导和转录激活因子3(signal transducers and activators of transcription3,STAT3)表达的影响。方法:70只雄性SD大鼠进行跑台递增负荷运动,采用Western blot测定安静时和急性运动后即刻、1h、3h、5h、12h、24h心肌CT-1以及p-STAT3/STAT3的表达。结果:运动后大鼠心肌CT-1表达量升高,其中运动后3h和5h时显著高于安静组(P<0.05),运动后12小时下降到运动前水平。运动后大鼠心肌p-STAT3/STAT3蛋白表达比值升高,其中运动后5h时显著高于安静组(P<0.05),运动后12h下降到运动前水平,运动后24h显著低于安静对照组(P<0.01)。结论:运动激活CT-1/STAT3通路,这种激活可能与运动中心肌功能代谢变化和运动诱发心肌肥大的发生有关。
Objective: To investigate the effect of exercise on the expression of cardiotrophin-1 (CT-1) and signal transducers and activators of transcription3 (STAT3) in rats. Methods: Seventy male Sprague-Dawley rats were subjected to treadmill exercise. The expressions of CT-1 and p-STAT3 / STAT3 in myocardium were detected by Western blot at rest and immediately after acute exercise, 1h, 3h, 5h, 12h and 24h. Results: The expression of CT-1 in myocardium of rats increased after exercise, especially at 3h and 5h after exercise (P <0.05), and decreased to pre-exercise 12 hours after exercise. After exercise, the ratio of p-STAT3 / STAT3 protein expression was increased in rats, which was significantly higher than that of sedentary group at 5h after exercise (P <0.05), decreased to pre-exercise level 12h after exercise and significantly lower than that of quiet control group 24h after exercise (P <0.01). CONCLUSION: Exercise activates CT-1 / STAT3 pathway, which may be related to functional changes in exercise-induced myocardial function and exercise-induced cardiac hypertrophy.
