玫瑰黄链霉菌(Streptomyces roseoflavus)Men-myco-93-63是分离自马铃薯疮痂病自然衰退土壤中的一株拮抗菌。该菌株及其发酵液对棉花黄萎病菌(Verticillium dahliae)等多种重要的植物病原真菌都具有较强的抑制作用。nsdA(negative regulator of Streptomyces differentiation)基因是从天蓝色链霉菌中发现的与抗生素合成相关的负调控基因。根据天蓝色链霉菌A3(2)的序列设计引物,PCR扩增出Men-myco-93-63nsdAmgh基因,经斑点杂交法验证,nsdAmgh确实在该生防菌中存在。该基因的发现有助于进一步了解该生防菌产生抗生素调节的分子机制,也为抗生素高产菌株的获得提供了新的靶标。
Streptomyces roseoflavus Men-myco-93-63 is an antagonistic bacterium isolated from naturally deteriorated soils of potato scab. The strain and its fermentation broth have a strong inhibitory effect on many important phytopathogenic fungi such as Verticillium dahliae. The negative regulator of Streptomyces differentiation gene is a negative regulator of antibiotic synthesis found in Streptomyces coelicolor. According to the sequence of Streptomyces coelicolor A3 (2), the gene of Men-myco-93-63nsdAmgh was amplified by PCR and confirmed by dot-blot hybridization that nsdAmgh indeed exists in this biocontrol strain. The discovery of this gene will help to further understand the molecular mechanism of antibiotic regulation of this biocontrol strain and also provide a new target for obtaining antibiotic-resistant high-yielding strains.
