目的:研究疏肝利胆颗粒对急性胰腺炎大鼠的保护作用。方法:SD大鼠随机分为正常组、模型组、疏肝利胆颗粒低剂量(3g/kg)组、中剂量(6g/kg)组、高剂量(12g/kg)组。灌胃给予相对应剂量的药物,每天1次连续灌胃给药10d,正常组与模型组给予等量的生理盐水。造模前禁食12h,自由饮水。末次灌胃给药后1h,除正常组外,其余各组大鼠每次按1.5g/kg腹腔注射6%左旋精氨酸(L-Arginine,L-Arg),共3次,每次间隔1h,诱导急性胰腺炎(acute pancreatitis,AP)大鼠模型。末次腹腔注射给药后12h处置动物,分光光度法测定一氧化氮(nitric oxide,NO)、一氧化氮合酶(nitric oxide synthase,NOS)、诱生性NO合成酶(inducible ni-tric oxide synthase,iNOS)、淀粉酶(amylase,AMS)的含量;放免法检测血清白细胞介素(interleukin,IL)-1和IL-6的含量;HE染色法检查胰腺组织病理学改变。结果:与模型组相比,疏肝利胆颗粒中、高剂量(6g/kg、12g/kg)组血清中NO、NOS、iNOS、AMS、IL-1和IL-6的含量明显降低,胰腺组织损伤程度明显减轻。结论:疏肝利胆颗粒对急性胰腺炎大鼠有一定的保护作用。
Objective: To study the protective effect of Shugan Lidan Granules on acute pancreatitis in rats. Methods: SD rats were randomly divided into normal group, model group, low dose group (3g / kg), medium dose (6g / kg) group and high dose (12g / kg) group. Gavage given the corresponding dose of drug, once a day continuous oral administration of 10d, the normal group and model group given the same amount of saline. Fasting 12h before modeling, free access to water. One hour after the last intragastric administration, rats in each group were injected intraperitoneally with 6% L-arginine (L-Arginine) at 1.5g / kg for 3 times except for the normal group, 1h, induced acute pancreatitis (AP) rat model. Animals were sacrificed 12h after the last intraperitoneal injection, and the nitric oxide (NO), nitric oxide synthase (NOS), inducible ni-tric oxide synthase iNOS, amylase (AMS) were detected by radioimmunoassay. Serum levels of interleukin (IL) -1 and IL-6 were detected by radioimmunoassay. Pathological changes of pancreas were examined by HE staining. Results: Compared with the model group, the content of NO, NOS, iNOS, AMS, IL-1 and IL-6 in the serum of ShuGanLiGan Granules were significantly decreased Tissue damage significantly reduced. Conclusion: Shugan Lidan Granule has a protective effect on acute pancreatitis in rats.
