为进一步探讨母胎免疫耐受的机理及妊娠和其相关疾病的免疫病因,对白细胞免疫球蛋白样受体1(LIR-1)在母胎界面的表达及其作用机制进行研究,发现LIR-1分子结构中含有免疫受体酪氨酸抑制基序,且分布广泛,在许多细胞上都被检测到表达,其通过与特异性MHC-Ⅰ类分子HLA-G及病毒U18蛋白等结合,传递负性信号,从而抑制靶细胞的活性及介导细胞因子的分泌等,在人类妊娠生理及病理妊娠如反复自然流产、妊娠期高血压疾病等中可能发挥极其重要的作用。
To further investigate the mechanism of immune tolerance in maternal-fetal and the immune etiology of pregnancy and its related diseases, we studied the expression and mechanism of leukocyte immunoglobulin-like receptor 1 (LIR-1) at maternal-fetal interface and found that LIR-1 The structure contains the immunoreceptor tyrosine inhibitory motif, which is widely distributed and has been detected on many cells. It binds to specific MHC class I molecules such as HLA-G and the viral U18 protein, delivering negative Signal, thereby inhibiting the activity of target cells and mediating the secretion of cytokines, which may play an extremely important role in physiological and pathological pregnancy of human beings such as recurrent spontaneous abortion and gestational hypertension.
