目的:研究黄管秦艽中龙胆苦苷对骨关节炎大鼠的治疗作用。方法:Wistar大鼠关节腔内注射造模剂造模,抽检确定造模成功后,将造模大鼠分为模型组、双醋瑞因(9 mg/kg)阳性对照组及龙胆苦苷低、中、高(50、100、200 mg/kg)剂量组,另设正常对照组,每组10只。各组大鼠灌胃给药,10 m L/kg。4 w后处死动物,取胸腺、脾脏,计算脏器指数;取血测全血粘度;制备血清测定IL-1β、PGE2、COX-2、COX-1、NO含量;摘取膝关节,制作病理切片HE染色观察组织病理学改变。结果:与正常对照组比较,模型组大鼠胸腺指数显著升高(P<0.05),全血低、中、高切粘度显著升高(P<0.01),血清IL-1β、PGE2、NO、COX-2含量显著升高(P<0.05或P<0.01);与模型组比较,各给药组胸腺指数显著降低(P<0.05),全血粘度显著降低(P<0.01),血清中IL-1β、PGE2、NO、COX-2含量显著降低(P<0.05或P<0.01);病理切片显示,与正常对照组比较,模型组关节软骨细胞排列紊乱且明显增生,染色严重降低,各给药组关节软骨细胞排列紊乱,染色中度降低。结论:龙胆苦苷可通过降低全血粘度及血清中IL-1β、PGE2、NO、COX-2含量起到对大鼠骨关节炎的治疗作用。
Objective: To study the therapeutic effect of gentiopicroside in the treatment of osteoarthritis in the yellow pipe. Methods: The Wistar rats were injected intraperitoneally with the model drug. Compared with the model group, the thymus index of each administration group decreased significantly (P <0.05), the whole blood viscosity decreased significantly (P <0.01) (P <0.05 or P <0.01). Conclusion: Gentiopicroside can reduce the whole blood viscosity and serum IL-1β, PGE2, NO, COX-2 levels play a role in the treatment of osteoarthritis in rats.
