目的:观察左归丸联合橄榄油对去势后大鼠骨组织和子宫的影响,探讨其防治绝经后骨质疏松症的机制。方法:对36只5~6月龄清洁型SD雌性大鼠随机分成4组:假手术组(Sham组),去卵巢组(OVX组),去卵巢+雌激素组(E2组),去卵巢+左归丸+橄榄油组(复方组)。用药方法:按1 ml/100g,每天1次灌胃,连续12周。对照组(①②组):每日以生理盐水灌胃。治疗组(③④组):E2组以己烯雌酚配成浓度为0.003 3 mg/ml的水溶液灌胃。复方组以中成药左归丸配成0.2g/ml生药的水溶液和初榨橄榄油交替每日灌胃1次。12周后分别左心室取血,检测血中E2、IL-1、IL-6水平,对腰椎及左侧股骨行双能X线骨密度测定。称子宫湿重并计算子宫系数,每组随机取2只行子宫组织切片观察。结果:OVX组中血清E2值明显低于Sham组(P<0.001),复方组中血清E2值分别与Sham组、E2组相比差异无统计学意义(P>0.05)。OVX组中IL-1与IL-6含量与其他3组相比均显著升高(P<0.05),E2组、复方组与Sham组间差异无统计学意义(P>0.05)。复方组的腰椎和左股骨骨密度较OVX组高,与Sham组差异均无统计学意义(P>0.05)。与Sham组比较,OVX组和复方组大鼠子宫湿重子宫系数下降(P<0.01);复方组的子宫湿重和子宫系数与OVX组比较无统计学差异(P>0.05)。子宫内膜HE染色镜下未见复方组子宫内膜增生现象,与E2组差异明显。结论:左归丸联合橄榄油能有效地减轻大鼠卵巢切除术引起的骨质丢失,且对子宫无或仅有轻微的刺激作用,机制可能是通过提高植物雌激素以调节炎症因子抑制破骨细胞的活性。
Objective: To observe the effect of Zuo Gui Pill combined with olive oil on bone tissue and uterus after castration in rats and to explore its mechanism of prevention and treatment of postmenopausal osteoporosis. Methods: Thirty-six SD female SD rats aged 5-6 months were randomly divided into 4 groups: Sham group, ovariectomized group (OVX group), ovariectomized + estrogen group (E2 group), ovariectomized + Zuogui Wan + olive oil group (compound group). Dual energy X-ray absorptiometry was performed on the lumbar spine and left femur. Weigh uterine wet weight and calculate the coefficient of the uterus, each group were taken 2 rows of uterine tissue sections were observed. Results: The serum E2 level in OVX group was significantly lower than that in Sham group (P <0.001). There was no significant difference in serum E2 between the two groups (P> 0.05) and Sham group and E2 group. The levels of IL-1 and IL-6 in OVX group were significantly higher than those in other three groups (P <0.05). There was no significant difference between E2 group, compound group and Sham group (P> 0.05). The BMD of lumbar and left femur of compound group was higher than that of OVX group, but there was no significant difference between Sham group and control group (P> 0.05). Compared with Sham group, the coefficient of uterine wet weight uterus in OVX group and compound group rats decreased (P <0.01). There was no significant difference in uterine wet weight and uterine coefficient in OVX group and OVX group (P> 0.05). There was no endometrial hyperplasia in compound group in HE staining, which was significantly different from E2 group. Conclusion: Zuogui Pill combined with olive oil can effectively reduce the bone loss caused by ovariectomy in rats, and it has no or only mild stimulating effect on the uterus. The mechanism may be that it inhibits osteoclasts by increasing phytoestrogen to regulate inflammatory cytokines Cell activity.
