研究1-(2,6-二甲基苯氧基)-2-(3,4-二甲基苯乙氨基)丙烷盐酸盐(DDPH)对心肌肥厚的逆转作用和对左室肌酸激酶(CK)及同工酶谱的影响。方法用部分狭窄腹主动脉的方法造成大鼠心肌肥厚模型,术后4w开始igDDPH25和50mg/kg·d-1,连续8w。结果术后12w,各组大鼠体重无显著性差异,但模型组左室重/全心重,全心重/体重分别比对照组高39.0%和36.9%,左室CK活性各组无明显差异,但模型组MB同工酶比对照组高3.4倍,MM同工酶降低143.6%.而2个给药组则明显使左室重/全心重和全心重/体重逆转,并减少肥厚心肌MB同工酶相对百分含量。结论DDPH可逆转部分狭窄腹主动脉所致心肌肥厚,同时可逆转心肌肥厚时肌酸激酶同工酶谱的变化。
To investigate the reversal effect of 1- (2,6-dimethylphenoxy) -2- (3,4-dimethylphenethylamino) propane hydrochloride (DDPH) on cardiac hypertrophy and the effects of left ventricular creatine kinase (CK) and isozyme patterns. Methods The rat model of myocardial hypertrophy was induced by partial narrowing of the abdominal aorta. IgDDPH25 and 50 mg / kg · d-1 started 4 w after the operation for 8 weeks. Results At 12 weeks after operation, there was no significant difference in body weight between the two groups. However, the left ventricular mass / whole heart weight and whole heart weight / body weight of the model group were 39.0% and 36.9% higher than that of the control group respectively. The left ventricular CK activity There was no significant difference in each group, but the model MB isozymes were 3.4 times higher than the control group, MM isozyme decreased 143.6%. However, the two administration groups obviously reversed left ventricular weight / total heart rate and total heart rate / body weight, and decreased the relative percentage of MB isozymes in hypertrophic myocardium. Conclusions DDPH can reverse the myocardial hypertrophy induced by stenosis of the abdominal aorta and reverse the changes of creatine kinase isoenzyme pattern during myocardial hypertrophy.
