目的研究自制方化湿液对湿阻证大鼠胃黏膜保护作用及对表皮生长因子受体(epithelial growth factor re-ceptor,EGFR)表达的影响。方法将40只SD大鼠分为正常组,模型组,化湿液低、中、高剂量组,每组8只。正常组常规喂养,其余各组采用改进的环境加疲劳法制造湿阻证模型,连续6 d。造模成功后正常组和模型组予生理盐水灌胃,化湿液治疗组按低、中、高剂量(4、8、16 g/kg)灌胃治疗,每天1次,连续8 d。采用比色法检测血清中褪黑素(MT)、谷胱甘肽过氧化物酶(GSH-Px)及丙二醛(MDA)的含量变化,并用免疫组化方法观察胃黏膜EGFR表达情况。结果与正常组相比,模型组大鼠脾虚症状明显,胃黏膜有明显充血水肿,并有出血点及血块,且血清MT及GSH-Px含量降低[MT:(9.68±1.48),GSH-Px:(368.10±52.92),P<0.05],MDA含量增高[(19.43±3.49),P<0.05];免疫组化显示,模型组大鼠胃黏膜EGFR表达较正常组减少[灰度值(166.013±7.757),光密度值(0.025±0.193),P<0.05]。经化湿液治疗后,与模型组比较,血清MT及GSH-Px含量增高(P<0.05),MDA含量降低(P<0.05),大鼠胃黏膜EGFR表达依次增加(P<0.05)。结论自制方化湿液可明显改善湿阻证大鼠脾虚症状并对胃黏膜损伤有保护作用,其机制可能与增强机体抗氧化应激能力及提高胃黏膜EGFR的表达量有关。
Objective To study the protective effect of self-made Chinese medicine on wet gastric mucosa and its effect on the expression of epidermal growth factor receptor (EGFR). Methods 40 SD rats were divided into normal group, model group and hygroscopic fluid low, medium and high dose group, 8 rats in each group. The rats in normal group were fed routinely, while the rest groups used the improved environment plus fatigue method to make the model of dampness resistance syndrome for 6 days. Normal group and model group were given gavage with normal saline after successful modeling. The rats in the wetting fluid group were treated with low, medium and high dose (4, 8, 16 g / kg) once a day for 8 days. Serum levels of melatonin (MT), glutathione peroxidase (GSH-Px) and malondialdehyde (MDA) were detected by colorimetric method. The expression of EGFR in gastric mucosa was observed by immunohistochemistry. Results Compared with the normal group, the symptoms of spleen deficiency in the model group were obvious. The gastric mucosa had obvious congestion and edema, bleeding spots and blood clots, and the levels of serum MT and GSH-Px were decreased [MT: (9.68 ± 1.48), GSH-Px (368.10 ± 52.92, P <0.05], MDA content increased (19.43 ± 3.49, P <0.05). Immunohistochemistry showed that the expression of EGFR in gastric mucosa of model group decreased ± 7.757), optical density (0.025 ± 0.193), P <0.05]. Compared with the model group, the levels of MT and GSH-Px in serum increased (P <0.05), the content of MDA decreased (P <0.05) and the expression of EGFR in gastric mucosa increased (P <0.05). CONCLUSION: Self-made Fanghua Huayu can obviously improve the symptoms of splenic asthenia in rats with dampness syndrome and protect the gastric mucosal injury. The mechanism may be related to the enhancement of antioxidant capacity and the expression of EGFR in gastric mucosa.
