利用定点突变技术获得的sIL-6R突变体H280Ⅰ基因与天然sIL-6R(wtsIL-6R)基因分别转染Cos7细胞,转染细胞上清在7TD1、R2细胞系上测定生物学活性,并同时进行IL-6结合能力分析。结果显示,sIL-6RH280Ⅰ可以保持同IL-6的结合能力,同时对IL-6的信号传递过程起拮抗作用,提示H280的改变可能影响IL-6R胞外区与gp130的结合。
The sIL-6R mutant H280Ⅰ gene and the natural sIL-6R (wtsIL-6R) gene obtained by site-directed mutagenesis were transfected into Cos7 cells, and the supernatant of transfected cells were assayed for biological activity on 7TD1 and R2 cell lines simultaneously IL-6 binding ability analysis. The results showed that sIL-6RH280Ⅰ could maintain the binding ability with IL-6 and antagonize the signal transduction of IL-6, suggesting that the change of H280 may affect the binding of IL-6R extracellular domain to gp130.
