以体外培养的人肺腺癌细胞SPC-A-1为材料,观察了β-胡萝卜素(0.1、1、10mg/L)对SPC-A-1细胞增殖的影响,以及它对SPC-A-1细胞DNA合成和细胞周期分布的影响。结果:台盼兰排斥试验表明,β-胡萝卜素对SPC-A-1细胞增殖有影响,且随β-胡萝卜素浓度增加和作用时间延长,对细胞的增殖的影响增大。当β-胡萝卜素浓度为0.1mg/L时,在用药后72h对SPC-A-1细胞才有明显抑制,(P<0.05),而在1、10mg/L浓度,于用药后48h就有明显的抑制作用(P<0.001)。~3H-TdR掺入SPC-A-1细胞DNA量的测定表明,不同浓度的β-胡萝卜素(0.1、1、10mg/L)对SPC-A-1细胞的DNA合成均有一定的抑制作用,抑制率分别为58.37%、59.66%、60.86%。与对照组有明显差别(P<0.05)。流式细胞仪(FCM)测定表明,24h时β-胡萝卜素对SPC-A-1细胞周期的分布影响不明显。
The effect of β-carotene (0.1, 1, 10 mg/L) on the proliferation of SPC-A-1 cells and its effect on SPC-A- 1 Effect of cellular DNA synthesis and cell cycle distribution. RESULTS: The trypan blue exclusion test showed that β-carotene had an effect on the proliferation of SPC-A-1 cells, and with the increase of β-carotene concentration and the prolonged action time, the effect on cell proliferation was increased. The DNA content of ~3H-TdR incorporation into SPC-A-1 cells showed that different concentrations of β-carotene (0.1, 1, 10 mg/L) had a certain inhibitory effect on DNA synthesis of SPC-A-1 cells. Flow cytometry (FCM) measurements showed that β-carotene had no significant effect on the distribution of SPC-A-1 cell cycle at 24 h.
