为观察p53基因和Rb基因与鼻咽癌发生的关系,应用聚合酶链反应-单链构型多态分析技术(PCR-SSCP技术),检测33例鼻咽癌组织、4例鼻咽部炎性组织及3例正常鼻咽部组织中p53基因第7,8外显子的突变。结果显示,在33例鼻咽癌中有7例存在p53基因第8外显子的突变,突变率为21.2%。在第7外显子中未检测出突变发生。应用Southern印迹杂交,对Rb基因在鼻咽癌中存在状态进行研究。结果表明,在13例鼻咽癌组织中有11例鼻咽癌组织有Rb基因的缺失或明显减弱,提示p53基因突变及Rb基因的缺失而导致的基因失活与鼻咽癌的发生有密切关系。
To observe the relationship between p53 gene and Rb gene and the occurrence of nasopharyngeal carcinoma, polymerase chain reaction-single strand conformation polymorphism (PCR-SSCP) was used to detect 33 cases of nasopharyngeal carcinoma, 4 cases of nasopharyngitis 3, and 3 normal nasopharyngeal tissues. The results showed that in 33 cases of nasopharyngeal carcinoma there are 7 cases of p53 gene exon 8 mutation, the mutation rate was 21.2%. No mutation was detected in exon 7. Southern blotting was used to study the existence of Rb gene in nasopharyngeal carcinoma. The results showed that in 13 cases of nasopharyngeal carcinoma, 11 cases of nasopharyngeal carcinoma have Rb gene deletion or significantly reduced, suggesting that p53 gene mutation and Rb gene deletion caused by gene inactivation and nasopharyngeal carcinoma are closely linked relationship.
