目的:探讨应激状态下胃粘膜耐受性细胞保护中转化生长因子α(TGFα)、表皮生长因子受体(EGFR)表达水平的变化。方法: 采用重复浸水束缚应激(WRS)制作动物模型;应用免疫组织化学方法动态检测TGFα及EGFR表达水平的变化。结果:TGFα主要位于正常粘膜增殖带细胞的胞质内,同时在胃腺基底部、主细胞及壁细胞上亦有表达,其表达部位较广泛;EGFR则定位于粘膜增殖带细胞的胞质及胞膜,以胞膜为主。单次应激使上述2 种细胞因子的表达均明显减弱,坏死区表达缺失。重复应激后,TGFα及EGFR的表达明显增强,而且除了粘膜增殖带外,在粘膜的其他部位(包括胃腺腔内)亦有较强表达。结论:胃粘膜耐受性细胞保护作用伴有TGFα及EGFR表达水平的增强,表明它们在这一现象中有重要的调节作用。
Objective: To investigate the changes of the expression of transforming growth factor-α (TGF-α) and epidermal growth factor receptor (EGFR) in gastric mucosal-tolerant cells under stress. Methods: Animal model was established by repeated immersion restraint stress (WRS). The expression of TGFα and EGFR was detected by immunohistochemical method. Results: TGFα mainly located in the cytoplasm of normal mucosa proliferating zone cells, and was also expressed on the base of gastric gland, major cells and parietal cells, with a wide range of expression sites. EGFR localized in the cytoplasm and cells Membrane, the membrane-based. A single stress so that the expression of the two cytokines were significantly weakened, the lack of expression of necrosis area. Repeated stress, TGFα and EGFR expression was significantly enhanced, and in addition to mucosal proliferation zone, in other parts of the mucosa (including gastric gland cavity) are also strongly expressed. CONCLUSION: Gastric mucosal tolerant cytoprotection accompanied with an increase in the expression of TGFα and EGFR suggests that they play an important regulatory role in this phenomenon.
