采用国际先进的AKTA-explorer蛋白快速纯化系统对蛇毒进行分离,通过多级分离与条件优化,将江浙蝮蛇毒镇痛组分分离纯化,并结合药理实验确定其镇痛组分。方法:江浙蝮蛇经过多步分离法分离。将分离的各组分,进行小鼠镇痛实验,以确定镇痛组分。结果:经三步柱分离可得单一色谱峰,纯度为99%。分离所得3、6号主峰具有显著镇痛作用。结论:利用三步柱分离法可将江浙蝮蛇毒中主要组分分离纯化。3、6号组分可基本确定为江浙蝮蛇毒镇痛组分。
The venom of snake venom was separated by the international advanced AKTA-explorer protein rapid purification system. Through multi-stage separation and optimization of conditions, the analgesic components of snake venom from Jiangsu and Zhejiang were isolated and purified, and their analgesic components were determined in combination with pharmacological experiments. Method: Jiangsu and Zhejiang snakes were separated by multiple step separation. The isolated components were subjected to analgesic experiments in mice to determine analgesic components. Results: A single chromatographic peak can be obtained by three-step column separation with a purity of 99%. The main peaks of No. 3 and No. 6 were isolated and had significant analgesic effect. Conclusion: The main components in snake venom of Jiangsu and Zhejiang can be separated and purified by three-step column separation. Components 3 and 6 can basically be identified as analgesic components of Jiangsu and Zhejiang snake venoms.
