人类常处于致癌环境中,但癌症的发病率仍维持在相对较低水平,这主要归功于内源性肿瘤抑制网络的存在。许多肿瘤抑制因子相继被发现,他们在细胞损伤后发挥的作用也越来越多的得到阐释。然而,是否存在一个通用靶基因来控制细胞存活与凋亡仍不十分清楚。为阻止基因突变并维持细胞稳定,需抑制这类靶基因表达。新近研究表明,凋亡抑制因子-Survivin可能是内源性肿瘤抑制网络的通用靶位,而Survivin沉默很可能是成功发挥肿瘤抑制作用的共同途径。本文将就Survivin及其通用靶位作用的研究进展做一综述。
Humans are often in a carcinogenic environment, but the incidence of cancer remains relatively low, largely due to the presence of an endogenous tumor suppressor network. Many tumor suppressors have been discovered one after another, and their role in cell injury is increasingly being elucidated. However, the existence of a universal target gene to control cell survival and apoptosis remains unclear. In order to prevent gene mutation and maintain cell stability, it is necessary to inhibit the expression of such target genes. Recent studies have shown that apoptosis-inhibitor Survivin may be a universal target for the endogenous tumor suppressor network, and silencing of Survivin is likely to be the common pathway for successful tumor suppression. This article will summarize the research progress of Survivin and its universal targets.
