目的分析药用菊花遗传多样性,为其种质资源评价和合理开发利用提供分子佐证。方法用ISSR分子标记对不同采集地的31份种源(菊花29份、野菊1份、菊花脑1份)进行遗传关系分析。结果22个引物共扩增出182个条带,其中149条具有多态性,多态位点百分率为81.87%,有效等位基因数(Ne)为1.348 1,Nei’s基因多样性指数(He)为0.219 1,Shannon信息指数(Ⅰ)为0.345 1。聚类分析和主坐标分析结果相近,将31份种源大致分为野菊和菊花脑、北方药用菊花、南方药用菊花3个类群。结论ISSR分子标记适合于药用菊花的品种鉴定和遗传多样性分析。
Objective To analyze the genetic diversity of medicinal chrysanthemum and provide molecular evidence for the evaluation of germplasm resources and their rational development and utilization. Methods ISSR molecular markers were used to analyze the genetic relationship among 31 provenances (29 chrysanthemums, 1 chrysanthemum and 1 chrysanthemum) from different habitats. RESULTS: A total of 182 bands were amplified by 22 primers, of which 149 were polymorphic, the percentage of polymorphic loci was 81.87%, the number of effective alleles (Ne) was 1.348 1, Nei’s gene diversity index (He) 0.2191 and Shannon Information Index (I) was 0.3451. Cluster analysis and the results of the main coordinate analysis are similar, the 31 provenances can be roughly divided into wild chrysanthemum and chrysanthemum brain, medicinal chrysanthemum in northern and southern medicinal chrysanthemum three groups. Conclusion ISSR molecular markers are suitable for the identification of medicinal chrysanthemum varieties and genetic diversity analysis.
