目的 探讨人参总皂甙对高能冲击波肾细胞凋亡相关基因的影响。方法 选用成年大耳白兔制备动物模型 ,将动物分为人参皂甙组、对照组 ,每组 8只 ,冲击波次数为 2 3 0 0~ 2 5 0 0次 ,工作电压 10 .5~ 12 .0kV。体外冲击波碎石 (ESWL)后 48h分别处死动物 ,取右肾行免疫组织化学链霉素抗生物素蛋白过氧化物酶连结法 (SP)染色。结果 bcl 2的表达 ,用药组肾曲管上皮阳性细胞数为 (87.6± 17.1)个 ,而对照组为 (2 2 .6± 8.2 )个 (P <0 .0 5 )。Bax表达 :用药组阳性细胞数为 (46.3± 11.7)个 ,对照组 (81.7± 9.6)个 (P <0 .0 5 )。结论 人参总皂甙可促使细胞凋亡抑制基因bcl 2高表达 ,相对抑制促进基因bax的表达 ,减少肾曲管上皮细胞凋亡
Objective To investigate the effects of ginsenosides on the apoptosis-related genes in high-energy shock wave-induced renal cells. Methods Adult large white rabbits were used to prepare animal models. The animals were divided into ginsenoside group and control group with 8 rats in each group. The number of shock wave was 2300 ~ 25000, the working voltage was 10.5 ~ 12.0kV . Animals were sacrificed 48 hours after ESWL, and the right kidney was stained with immunohistochemical streptavidin-peroxidase-linked immunosorbent assay (SP). Results The expression of bcl 2 was significantly higher in the experimental group than in the untreated group (87.6 ± 17.1), compared with (22.6 ± 8.2) in the control group (P <0.05). Bax expression: The number of positive cells in the treatment group was (46.3 ± 11.7) and in the control group (81.7 ± 9.6) (P <0.05). Conclusion Ginseng total saponin can promote the expression of apoptosis-inhibiting gene bcl 2, inhibit the expression of gene bax and decrease the apoptosis of renal tubule epithelial cells
