富血小板血浆可注射组织工程骨促进兔下颌牵张成骨实验研究

目的探讨富血小板血浆可注射组织工程骨对兔下颌骨牵张成骨新骨形成的促进作用。方法将40只新西兰白兔随机分为A组、B组、C组、D组各10只。建立下颌骨牵张成骨动物模型,固定期第2天,A组于牵张间隙注射富血小板血浆可注射组织工程骨,B组注射富血小板血浆和纤维蛋白胶原的混合液,C组注射纤维蛋白胶原,D组注射生理盐水作为空白对照组。分别于固定2、6周时处死动物获取标本,通过大体观察、HE染色、X线观察、骨密度分析观察骨质愈合改建情况。数据进行单因素方差分析和组间两两比较的SNK-q检验,P<0.05为差异有统计学意义。结果通过大体观察、HE染色、X线观察、骨密度分析显示,在固定期第2、6周,A组、B组牵张间隙内成骨质量好于C组、D组,A组优于B组。A组固定第2、6周牵张区新生骨骨密度值分别为(66.74±1.97)、(94.65±2.35)g/cm2,B组分别为(43.63±1.84)、(68.54±2.03)g/cm2,C组分别为(27.15±1.49)、(53.69±1.64)g/cm2,D组分别为(25.39±1.23)、(55.32±1.52)g/cm2,A组牵张区内新生骨骨密度明显高于B组、C组、D组(均P<0.05),B组明显高于C组和D组(均P<0.05)。结论富血小板血浆可注射组织工程骨可有效促进兔下颌骨牵张成骨过程中的新骨形成。 Objective To investigate the promoting effect of platelet-rich plasma injectable tissue engineering bone on new bone formation in rabbit mandibular distraction osteogenesis. Methods Forty New Zealand White rabbits were randomly divided into A group, B group, C group and D group. The animal model of mandibular distraction osteogenesis was established. On the second day of fixation, group A was injected with platelet-rich plasma into the interstitial space to inject tissue engineering bone. Group B was injected with a mixture of platelet-rich plasma and fibrin collagen. Collagen, Group D was injected with normal saline as blank control group. The animals were sacrificed at 2 and 6 weeks respectively to obtain the specimens. The changes of bone remodeling were observed by gross observation, HE staining, X-ray and bone mineral density analysis. Data were analyzed by one-way ANOVA and SNK-q test between groups, P <0.05 was considered statistically significant. Results Gross observation, HE staining, X-ray observation and bone mineral density analysis showed that in the 2nd and 6th week of fixation, the osteogenetic quality of group A and group B was better than that of group C, while group D and group A were better than group C Group B. In group A, the newborn bone densities in the distracted areas were (66.74 ± 1.97) and (94.65 ± 2.35) g / cm2 at the 2nd and 6th week respectively, and those in the B group were (43.63 ± 1.84) and (68.54 ± 2.03) g / cm2 in group A were (27.15 ± 1.49) and (53.69 ± 1.64) g / cm2 respectively in group C and (25.39 ± 1.23) and (55.32 ± 1.52) g / cm2 in group D. The bone mineral density Which were significantly higher than those in group B, group C and group D (all P <0.05), group B was significantly higher than group C and group D (all P <0.05). Conclusion Platelet-rich plasma injected with tissue engineered bone can effectively promote new bone formation during mandibular distraction osteogenesis in rabbits.
