IL-12是一种具有广泛免疫调节作用和抗肿瘤作用的细胞因子,研究了IL-12对白介素4基因转染的小鼠B16黑色素瘤细胞(Bl6-IL-4 ̄+)诱导的CTL的杀伤活性、IFN-γ分泌水平和增殖能力的影响。结果发现,IL-12能显著增强体外诱导的CTL的杀伤活性及提高IFN-γ的分泌水平,并能促进CTL的增殖。IL-12与低剂量的IL-2(50U/ml)合用能协同增强CTL的杀伤活性、IFN-γ分泌水平及增殖能力, 但与高剂量的IL-2(500U/ml)合用却降低了促CTL诱导作用,结果表明,IL-12 作为瘤苗增强剂可望促进肿瘤细胞靶向的IL-4基因治疗的效果,与低剂量的IL-2合用后效果更佳。
