目的探讨microRNA let-7i调控树突状细胞(DC)功能的作用靶点。方法分别上调或下调DC中let-7i水平,应用双萤光素酶报告基因系统检测SOCS1是否是let-7i的作用靶点,应用Western blot及免疫荧光检测DC中SOCS1蛋白水平,qRTPCR检测DC中SOCS1及let-7i基因水平,观察在调控DC功能的过程中,let-7i是否靶向抑制SOCS1表达。结果 let-7i通过转录后抑制的方式靶向调控DC中SOCS1表达。结论 miRNA let-7i靶向抑制SOCS1,进而影响DC的成熟及功能状态。
Objective To investigate the role of microRNA let-7i in regulating the function of dendritic cells (DCs). Methods The levels of let-7i in DCs were up-regulated or down-regulated respectively. Whether or not SOCS1 was the target of let-7i was detected by dual luciferase reporter system. Western blot and immunofluorescence were used to detect the level of SOCS1 protein in DCs. SOCS1 and let-7i gene levels, to observe whether let-7i inhibits SOCS1 expression during the regulation of DC function. As a result, let-7i targeted SOCS1 expression in DCs by means of post-transcriptional inhibition. Conclusions miRNA let-7i targets to inhibit SOCS1, thereby affecting the maturation and functional status of DCs.
