目的:建立高效毛细管电泳法同时测定不同产地多叶棘豆中3种黄酮类化合物含量的方法。方法:采用高效毛细管电泳法,弹性石英毛细管柱75μm×50 cm,电动进样15 s,分离电压18 kV,毛细管温度25℃,检测波长为283 nm,运行缓冲液为0.25 mol.L-1硼砂30%乙腈溶液。结果:不同产地多叶棘豆中3种黄酮类化合物的含量有较大差异,所用实验条件下,3种对照品溶液分别在1.00~40.00(r=0.999 8),2.00~80.00(r=0.999 9),0.50~80.00 mg.L-1(r=0.999 6)具有良好的线性关系,平均回收率均>96.00%,RSD<2.0%。结论:建立的高效毛细管电泳法可作为多叶棘豆质量控制的一种方法。
OBJECTIVE: To establish a method for the simultaneous determination of three flavonoids in Acanthostris from different habitats by high performance capillary electrophoresis. Methods: High-performance capillary electrophoresis was used. The elastic quartz capillary column was 75μm × 50 cm, electrokinetic injection was performed for 15 seconds, the separation voltage was 18 kV, the capillary temperature was 25 ℃, the detection wavelength was 283 nm and the running buffer was 0.25 mol·L-1 borax 30% acetonitrile solution. Results: The contents of three flavonoids in different origins of Oxytropis were significantly different. Under the experimental conditions, the concentrations of three reference solutions were 1.00-40.00 (r = 0.999 8), 2.00-80.00 (r = 0.999 9), 0.50 ~ 80.00 mg.L-1 (r = 0.999 6). The average recoveries were all> 96.00% with RSD <2.0%. Conclusion: The established high performance capillary electrophoresis can be used as a quality control method of Eupatorium.
