目的研究环孢菌素-A(Cs A)对体外培养的大鼠树突状细胞前体(DCp)的表型及功能的影响。方法在大鼠骨髓来源的DC前体培养过程中加入CsA(1 mg/L),并设对照组,培养期间观察DC形态,流式细胞仪分析细胞表型,混合淋巴细胞反应检测其刺激同种T细胞增殖能力。结果CSA显著抑制了DCp的成熟过程,抑制其表面共刺激分子的表达,并显著抑制其刺激同种T细胞增殖能力。结论CsA可抑制DC成熟,使DC处于未成熟状态,从而发挥免疫抑制作用。
Objective To study the effect of cyclosporine-A (Cs A) on the phenotype and function of dendritic cell precursor (DCp) cultured in vitro. Methods CsA (1 mg / L) was added to the bone marrow-derived DC precursors of rats and the control group was established. The morphology of DCs was observed during the culture. The phenotype of cells was analyzed by flow cytometry. The mixed lymphocyte reaction T cell proliferation ability. Results CSA significantly inhibited the maturation of DCp, inhibited the expression of co-stimulatory molecules on the surface and significantly inhibited its ability to stimulate allogeneic T cell proliferation. Conclusions CsA can inhibit the maturation of DC and make the DCs immature and thus play an immunosuppressive role.
