目的:明确解毒化瘀颗粒在急性肝衰竭动物实验中的治疗作用及是否可抑制急性肝衰竭大鼠肝线粒体通透性转换。方法:采用D氨基半乳糖联合内毒素脂多糖构建急性肝衰竭大鼠模型,解毒化瘀颗粒干预治疗,成模后24 h收集各组大鼠标本,同时观察各组大鼠48 h存活率,全自动生化仪检测血清ALT、AST含量,分光光度仪检测肝线粒体在520 nm处吸光度的变化来测定PT孔开放情况,Western Bolt检测肝线粒体与胞质细胞色素C蛋白含量,测定Caspase-3蛋白活性情况。结果:解毒化瘀颗粒可提高急性肝衰竭大鼠48 h存活率,降低血清ALT、AST含量,抑制肝线粒体PT孔开放,提高线粒体细胞色素C含量,降低细胞质细胞色素C含量,抑制下游Caspase-3蛋白活性。结论:解毒化瘀颗粒可抑制急性肝衰竭大鼠肝线粒体通透性转换,抗急性肝衰竭肝细胞凋亡。
Objective: To clarify the therapeutic effect of Jiedu Huayu Granule in animal experiments of acute hepatic failure and whether it can inhibit hepatic mitochondrial permeability transition in rats with acute hepatic failure. Methods: A rat model of acute hepatic failure was established by D-galactosamine combined with lipopolysaccharide (LPS), and the treatment of Jiedu Huayu granule was taken. The rats were collected at 24 h after the model was established. At the same time, The contents of ALT and AST in serum were detected by automatic biochemical analyzer. The openness of PT pores was measured by the change of absorbance at 520 nm by spectrophotometer. The contents of mitochondria and cytoplasmic cytochrome C in liver were detected by Western Bolt. The expressions of Caspase-3 protein Active situation. Results: Jiedu Huayu Granule can improve the survival rate of rats with acute hepatic failure at 48 h, decrease the content of ALT and AST in serum, inhibit the opening of PT of hepatic mitochondria, increase the content of cytochrome C in mitochondria, decrease the content of cytochrome C in the mitochondria and inhibit the expression of Caspase- 3 protein activity. Conclusion: Jiedu Huayu Granule can inhibit hepatic mitochondrial permeability transition in acute liver failure rats and prevent hepatocellular apoptosis in acute liver failure.
