目的:观察大鼠视神经夹伤后视网膜中GAP-43和Nogo-A蛋白的动态变化,探讨左归丸对大鼠视神经部分损伤后再生修复的影响。方法:建立大鼠单侧视神经部分损伤模型,分别给予左归丸液(实验组,2.0g.kg-1.d-1)和等量生理盐水(对照组)灌胃,采用免疫组织化学技术于伤后不同时间点检测GAP-43及Nogo-A蛋白在受损眼视网膜中的表达。结果:视神经损伤后,GAP-43和Nogo-A蛋白表达先升高后下降,伤后2~4周时左归丸组GAP-43表达显著上调,而Nogo-A表达基本降至正常水平,与对照组相比,差异有显著性(P<0.01)。结论:左归丸可显著促进GAP-43的表达,同时抑制Nogo-A表达,促进受损视神经再生修复。
OBJECTIVE: To observe the dynamic changes of GAP-43 and Nogo-A protein in the retina after optic nerve crush in rats and explore the effect of Zuogui Pill on the regeneration and repair of partial optic nerve injury in rats. Methods: A rat model of unilateral optic nerve partial injury was established. Zuogui pill solution (2.0g.kg-1.d-1) and saline (control group) were intragastrically administrated. Immunohistochemistry At different time points after injury, the expression of GAP-43 and Nogo-A protein in damaged retina was detected. Results: After optic nerve injury, the expression of GAP-43 and Nogo-A protein increased firstly and then decreased, and the expression of GAP-43 in Zuoguiwan group increased significantly at 2 ~ 4 weeks after injury, while the expression of Nogo-A decreased to normal level, Compared with the control group, the difference was significant (P <0.01). Conclusion: Zuogui pill can significantly promote the expression of GAP-43, inhibit the expression of Nogo-A and promote the regeneration of damaged optic nerve.
